【摘要】：【目的】研究并確定耐熱保護(hù)劑在仔豬副傷寒活疫苗中應(yīng)用的各參數(shù),為制備細(xì)菌耐熱保護(hù)劑活疫苗提供參考�！痉椒ā繉⒛蜔岜Ｗo(hù)劑與制苗菌液等體積混合,分別采用3種常用凍干曲線(1、2、3)進(jìn)行凍干,抽樣檢測(cè)確定耐熱保護(hù)劑配套凍干曲線。在確定合適凍干曲線的基礎(chǔ)上,進(jìn)一步研究耐熱保護(hù)劑與不同菌齡菌液(發(fā)酵培養(yǎng)12、15、18和21 h)、不同濃度菌液(菌液終濃度分別為3×1010、5×1010和7×1010 CFU/m L)、不同感作時(shí)間(分別作用0、24和48 h)、耐熱保護(hù)劑的不同保存時(shí)間(2-8°C分別保存0、10、20、30和40 d),以及不同分裝量(2、3和4 m L)等多種不同參數(shù)對(duì)疫苗質(zhì)量的影響�！窘Y(jié)果】配套凍干曲線研究表明,曲線2凍干的產(chǎn)品性狀、活菌存活率與耐老化指標(biāo)效果最好;菌齡研究表明,37°C發(fā)酵培養(yǎng)18 h(位于對(duì)數(shù)生長(zhǎng)末期或穩(wěn)定前期),其凍干菌存活率達(dá)78%-81%,優(yōu)于其它時(shí)間;配苗試驗(yàn)表明,菌液適宜終濃度為3×1010-5×1010 CFU/m L;最適感作時(shí)間為2-8°C感作24 h,凍干菌存活率可達(dá)85.3%-90.5%;耐熱保護(hù)劑保存期試驗(yàn)表明,2-8°C保存40 d與0 d(配制當(dāng)日)的保護(hù)劑凍干效果基本一致;配苗分裝量試驗(yàn)表明,7 m L西林瓶中分裝3 m L或4 m L,其凍干菌存活率與2 m L基本一致,但耐老化試驗(yàn)中活菌存活率比2 m L略高�！窘Y(jié)論】收獲發(fā)酵培養(yǎng)18 h的菌液、配苗終濃度采用3×1010-5×1010 CFU/m L,使用2-8°C保存40 d內(nèi)的耐熱保護(hù)劑,讓保護(hù)劑與制苗用菌液2-8°C感作24 h,采用3-4 m L進(jìn)行定量分裝,按曲線2凍干為制備仔豬副傷寒耐熱保護(hù)劑活疫苗的適合參數(shù)。
[Abstract]:[objective] to study and determine the parameters of the application of heat-resistant protective agent in live vaccine of piglet paratyphoid fever, and to provide a reference for the preparation of live vaccine of bacterial heat-resistant protectant. Three kinds of freeze-drying curves (1 ~ 2 ~ 3 ~ 3) were used for freeze-drying, and the freeze-drying curves of heat-resistant protectant were determined by sampling test. On the basis of determining the suitable freeze-drying curve, the heat resistance protectant and the bacteria solution of different ages were further studied. The final concentration of the bacterial solution was 3 脳 10 10 5 脳 1010 and 7 脳 1010 CFU/m L), respectively. The results of seedling matching test showed that the survival rate of lyophilized bacteria was basically the same as that of 2 mL. But the survival rate of live bacteria in aging tolerance test was slightly higher than that in 2 mL. [conclusion] the final concentration of seedling was 3 脳 10 ~ (-5) 脳 1010 CFU/m / L and stored at 2-8 擄C for 40 days. For 24 h, the protective agent and the inoculation liquid were divided quantitatively with 3-4 mL, and freeze-dried according to curve 2 was used as the suitable parameter for the preparation of live vaccine of paratyphoid heat resistance protective agent in piglets.
