益生菌對柔嫩艾美爾球蟲感染雛雞免疫水平的影響及免疫保護(hù)效果評價

【摘要】:雞球蟲病呈全球性分布,在養(yǎng)禽業(yè)中普遍存在且危害極大,往往造成巨大的經(jīng)濟(jì)損失且嚴(yán)重阻礙養(yǎng)禽業(yè)發(fā)展。益生菌有望成為防治球蟲病的一條新路徑。由于益生菌的指代范圍極其廣泛,所以本研究以乳酸菌為例探究益生菌對柔嫩艾美爾球蟲(E.tenella)感染雛雞免疫水平的影響及其免疫保護(hù)效果。本研究以1~31日齡雄性肉雛雞為實驗動物,并選取了3種乳酸菌菌株進(jìn)行試驗,試驗選用的3種乳酸菌菌株分別為鼠李糖乳桿菌(Lactobacillus rhamnosus)ATCC53103、乳酸片球菌(Pediococcus acidilactici)ATCC8042和植物乳桿菌(Lactobacillus plantarum)WCFS1。將體重相近,健康的250只1日齡肉用雛雞隨機(jī)分為A、C、D、E 5組,每組50只分別飼養(yǎng)于經(jīng)嚴(yán)格消毒且無球蟲感染的環(huán)境中。雛雞1~21日齡期間,A、B兩組每日飼喂前空腹灌服生理鹽水200μL/只,C、D、E三組每日飼喂前空腹灌服各自相應(yīng)菌種2×108cfu/200μL,其中C組雛雞灌服Lactobacillus rhamnosus ATCC53103,D組雛雞灌服Pediococcus acidilactici ATCC8042,E組雛雞灌服Lactobacillus plantarum WCFS1。雛雞22日齡時,每組處死25只進(jìn)行外周血中淋巴細(xì)胞增殖能力的檢測、CD3+CD4+和CD3+CD8+T淋巴細(xì)胞含量的檢測、細(xì)胞因子IL-2和IFN-γ分泌量的檢測、抗體Ig G含量的檢測以及食糜中SIgA含量的檢測。除A組外其它各組余下雛雞分別經(jīng)口感染已孢子化的E.tenella卵囊5×104個/只,攻蟲8天后,雛雞31日齡時處死全部雛雞,除檢測上述免疫指標(biāo)外,對雛雞的相對增重率、血便記分、盲腸病變程度、盲腸內(nèi)容物中的卵囊值、糞便中的卵囊排出量、抗球蟲指數(shù)及病理組織切片也進(jìn)行相應(yīng)檢測。具體試驗結(jié)果如下:(1)乳酸菌對E.tenella感染雛雞免疫水平的影響:球蟲感染前、后,雛雞22日齡和31日齡時分別對雛雞進(jìn)行相關(guān)免疫指標(biāo)檢測,結(jié)果表明試驗所用三株乳酸菌在球蟲感染前、后均能夠提高雛雞外周血中淋巴細(xì)胞的增殖能力,促進(jìn)外周血中CD3+CD4+和CD3+CD8+T淋巴細(xì)胞的生成,進(jìn)而提高外周血中細(xì)胞因子IL-2和IFN-γ的分泌量,而提高雛雞細(xì)胞免疫水平;另外,試驗所用三株乳酸菌也能夠通過提高外周血中IgG抗體及腸內(nèi)容物中SIgA抗體的分泌量從而提高雛雞體液免疫水平。其中免疫保護(hù)作用效果最好的菌株是Lactobacillus rhamnosus ATCC53103,Lactobacillus plantarum WCFS1次之,Pediococcus acidilactici ATCC8042最次。(2)乳酸菌對雞E.tenella的免疫保護(hù)效果評價:結(jié)果表明在球蟲感染前、后,灌服乳酸菌的3組雛雞其平均增重均比B組高,且C組和B相比差異顯著(P㩳0.05);灌服乳酸菌的C、D、E三組雛雞其血便記分均低于B組,且C組最低;B組雛雞盲腸病變最嚴(yán)重,病變記分平均值達(dá)到3.25,極顯著的高于C、E兩組(P㩳0.01),顯著的高于D組(P㩳0.05);灌服乳酸菌的C、D、E三組雛雞其卵囊排出量均低于B組,且C組最低;灌服乳酸菌的三組雛雞其抗球蟲指數(shù)分別為152.40、123.22、133.26,均在120~160之間,具有中等的抗球蟲效果,且Lactobacillus rhamnosusATCC53103的抗球蟲效果最好,Lactobacillus plantarum WCFS1次之,Pediococcus acidilactici ATCC8042最次。
[Abstract]:Cocciasis is a global distribution, widespread and harmful in poultry industry, which often leads to huge economic losses and seriously hinders the development of poultry industry. Probiotics are expected to be a new path to the prevention and treatment of coccidia. In this study, lactic acid bacteria were used as an example to investigate the effect of probiotics on the immune level of chicken infected with Eimeria tenella (E.tenella) and its immune protective effect. Three lactic acid bacteria strains were selected as experimental animals and three kinds of lactic acid bacteria strains were selected. The three lactic acid bacteria strains selected were Lactobacillus rhamnosus ATCC 53103, coccus acidilactici ATCC 8042 and Lactobacillus plantarum WCFS1, respectively. The broiler chicks were randomly divided into A, C, D and E 5 groups, and 50 animals in each group were housed in an environment of strict disinfection and no coccidia infection. During the age of 1-21 days, two groups of groups A and B were fed with normal saline (200. mu.L/ L, C, D, E) before daily feeding, and the two groups were fed with the corresponding strain of 2 ~ 108cfu/ 200 & mu; L. Group E chicks were inoculated with Lactobacillus plantarum WCFS1. The detection of lymphocyte proliferation in peripheral blood, the detection of CD3 + CD4 + and CD3 + CD8 + T lymphocyte content, the detection of cytokine IL-2 and IFN-IL secretion, the detection of antibody Ig G content and the detection of SIgA content in chyme were carried out at the age of 22 days. Except for the above-mentioned immunological indexes, the relative weight gain, blood stool score and cecum lesion degree of chicks were determined. The egg sac value in the cecal contents, the amount of egg sac discharge in the feces, the anticoccidial index and the pathological tissue sections were also detected accordingly. The specific test results are as follows: (1) The effect of lactic acid bacteria on immune level of chickens infected with E.tenella is as follows: before and after the infection of coccidia, the chickens are tested with relevant immune indexes at the age of 22 days and the age of 31 days respectively, and the results show that the three lactic acid bacteria used in the test are before the infection of coccidian. The method can improve the proliferation ability of the lymphocytes in the peripheral blood of the chicken, promote the generation of CD3 + CD4 + and CD3 + CD8 + T lymphocytes in the peripheral blood, and further improve the secretion of the cytokine IL-2 and IFN-IL in the peripheral blood, thereby improving the immune level of the chicken cells; and additionally, The three lactic acid bacteria used in the test can also improve the humoral immunity level of the chicken by increasing the secretion amount of the SIgA antibody in the IgG antibody and the intestinal contents in the peripheral blood. Among them, the best strain was Lactobacillus rhamnosus ATCC 53103, Lactobacillus plantarum WCFS1 was the second, and gioctactici ATCC 8042 was the best. (2) The effect of lactic acid bacteria on the immune protection of chicken E. Lacella showed that the average weight gain was higher than that of group B before and after coccidia infection, and the difference between group C and B was significantly higher than that in group B (P? 0.05); C, D of lactic acid bacteria were perfused with lactic acid bacteria. The scores of blood stool were lower in group E than in group B, and the lowest in group C, and the average value of lesion score was 3. 25 in group B, which was significantly higher than that of group C and E (P? 0.01), which was significantly higher than that of group D (P? 0.05); C, D of lactic acid bacteria in group B were significantly higher than that in group D (P? 0.05). The number of egg sacs was lower in group E than in group B, and the lowest in C group. The anticoccidia index of the three groups with lactic acid bacteria was 152. 40, 123. 22, 133. 26, all of which were 120 ~ 160, had moderate anti-coccidia effect, and Lactiobacter rhamnosous 53103 had the best anti-coccidial effect, and Lactobacillus plantarum WCFS1 was the second.
