IDO在梅花鹿茸角中的表達(dá)及對鹿茸軟骨細(xì)胞分化的影響

【摘要】:以梅花鹿茸角為研究對象,通過原位雜交方法檢測吲哚胺2,3-雙加氧酶1(IDO1)和吲哚胺2,3-雙加氧酶2(IDO2)在茸角中的表達(dá)。在培養(yǎng)的鹿茸軟骨細(xì)胞中分別添加IDO1抑制劑1-甲基-L-色氨酸(1-L-MT)和IDO2抑制劑1-甲基-D-色氨酸(1-D-MT),作用24h后,通過熒光定量PCR方法檢測軟骨細(xì)胞分化標(biāo)志分子Ⅹ型膠原(ColⅩ)表達(dá)的變化,進(jìn)一步研究IDO1和IDO2對鹿茸軟骨細(xì)胞分化的影響。結(jié)果顯示:IDO1和IDO2主要在鹿茸真皮層成纖維細(xì)胞、間充質(zhì)細(xì)胞和軟骨細(xì)胞中表達(dá)。用濃度分別為200μmol/L和100μmol/L的1-L-MT和1-DMT處理鹿茸軟骨細(xì)胞24h后,熒光定量PCR結(jié)果顯示,ColⅩmRNA在鹿茸軟骨細(xì)胞中的表達(dá)量均顯著下降。結(jié)果表明:當(dāng)IDO的活性受到抑制時,鹿茸軟骨細(xì)胞的分化過程也受到明顯抑制,IDO可能在梅花鹿鹿茸軟骨細(xì)胞分化過程中起重要的作用。
[Abstract]:The expression of indoleamine (IDO1) and indole amine (IDO2) in antler horn were detected by in situ hybridization in the antler of Sika deer antler. The results showed that the expression of indoleamine 3-dioxygenase 1 (IDO1) and indoleamine 3-dioxygenase 2 (IDO2) in antler horn were detected by in situ hybridization. Cultured velvet chondrocytes were treated with IDO1 inhibitor 1-methyl-L-tryptophan (1-L-MT) and IDO2 inhibitor 1-methyl-Dtryptophan (1-D-MT) for 24 h.
