【摘要】：為了研究反芻動(dòng)物瘤胃液內(nèi)各有機(jī)酸含量水平,建立同時(shí)測(cè)定瘤胃液中乳酸和6種揮發(fā)性脂肪酸的高效液相色譜分析方法,試驗(yàn)采用離子排斥色譜模式,采用Sepax Carbomix H-NP 5(300×7.8 mm ID,5μm,8%交聯(lián)度)色譜柱,配同系保護(hù)柱,柱溫55℃,流動(dòng)相采用2.5 mmol/L H2SO4水溶液,流速0.50 m L/min,示差折光檢測(cè)器溫度30℃,響應(yīng)時(shí)間6 s。結(jié)果表明:乳酸和6種揮發(fā)性脂肪酸30 min內(nèi)基線分離無干擾,在5.0~200.0 mg/L濃度范圍內(nèi)呈現(xiàn)良好的線性關(guān)系,各組分方法檢測(cè)限在0.24~0.46 mg/L之間。說明該方法樣品前處理簡(jiǎn)便,重現(xiàn)性好,結(jié)果準(zhǔn)確。
