豬線粒體抗病毒信號蛋白MAVS對口蹄疫病毒感染宿主的影響

【摘要】:采用構(gòu)建豬源MAVS重組質(zhì)粒pEGFP-MAVS,將其轉(zhuǎn)染PK-15細(xì)胞24h后,再感染口蹄疫病毒(MOI=0.1),通過Western blot和real-time PCR檢測MAVS融合蛋白表達及對口蹄疫病毒感染宿主細(xì)胞的復(fù)制和病毒感染滴度。結(jié)果顯示:重組質(zhì)粒pEGFP-MAVS能在PK-15細(xì)胞中獲得表達,并且上調(diào)MAVS基因?qū)谔阋卟《驹谒拗骷?xì)胞中的早期復(fù)制有顯著抑制作用,病毒的感染滴度也有一定的降低。結(jié)果表明:MAVS具有一定的抗病毒作用,此為進一步研究口蹄疫病毒逃逸宿主天然免疫的機制奠定了基礎(chǔ)。
[Abstract]:The porcine MAVS recombinant plasmid pEGFP-MAVS, was constructed and transfected into PK-15 cells for 24 hours and then infected with foot-and-mouth disease virus (MOI=0.1). The expression of MAVS fusion protein and the replication and titer of FMDV infected host cells were detected by Western blot and real-time PCR. The results showed that the recombinant plasmid pEGFP-MAVS could be expressed in PK-15 cells, and upregulation of MAVS gene could significantly inhibit the early replication of FMDV in host cells, and the infection titer of FMDV was also decreased to some extent. The results showed that WMAVS had a certain antiviral effect, which laid a foundation for further study on the mechanism of innate immunity of foot-and-mouth disease virus escape host.
