飼用優(yōu)良乳酸菌鑒定及生物學(xué)特性研究

【摘要】:對(duì)9株供試乳酸菌產(chǎn)酸產(chǎn)氣、生長速率、產(chǎn)酸速率等方面篩選既能用于微生態(tài)制劑又能用于青貯飼料發(fā)酵劑的優(yōu)良乳酸菌;并從細(xì)菌形態(tài)學(xué)、生理生化特征和16SrDNA基因序列分析對(duì)其進(jìn)行鑒定;同時(shí)進(jìn)一步研究乳酸菌對(duì)溫度、鹽濃度和強(qiáng)酸強(qiáng)堿的耐受性、有機(jī)酸組分和藥物敏感性等生物學(xué)特性。試驗(yàn)篩選出植物乳桿菌R02(Lactobacillus plantarum)、糞腸球菌R07(Enterococcus faecalis)和戊糖片球菌R09(Pediococcus pentosaceus)3株優(yōu)良乳酸菌。其中菌株R09生長最快,產(chǎn)酸能力最強(qiáng),乳酸占總酸含量最高,達(dá)95.45%,可耐受0.12g/mL NaCl;R02對(duì)溫度的耐受性最強(qiáng),可耐受55℃高溫;R07對(duì)強(qiáng)酸強(qiáng)堿環(huán)境有較強(qiáng)的耐受能力,在pH為3.0~9.5的條件下均可生長。菌株R02、R07、R09均具有生長快、產(chǎn)酸和耐受能力強(qiáng)的優(yōu)良生物學(xué)特性,因此具有應(yīng)用于微生態(tài)制劑和青貯飼料的潛能。
[Abstract]:In this paper, 9 strains of lactic acid bacteria which can be used in both microecological preparation and silage starter were screened from the aspects of gas production, growth rate and acid production rate of lactic acid bacteria, and the morphology of bacteria was analyzed. Physiological and biochemical characteristics and 16SrDNA gene sequence analysis were used to identify lactic acid bacteria. The biological characteristics of lactic acid bacteria such as temperature, salt concentration, tolerance to strong acid and alkali, organic acid composition and drug sensitivity were also studied. Three strains of Lactobacillus plantarum R02 (Lactobacillus plantarum), Enterococcus faecalis R07 (Enterococcus faecalis) and Streptococcus pentose R09 (Pediococcus pentosaceus) were screened out. The strain R09 has the fastest growth, the strongest acid production ability, the highest lactic acid content, up to 95.455.The strain R09 can tolerate the highest temperature of 0.12g/mL NaClCon R02, and can tolerate 55 鈩,
