復(fù)合菌發(fā)酵玉米蛋白粉的條件優(yōu)化

【摘要】:試驗旨在研究復(fù)合菌(黑曲霉、乳酸菌、酵母菌和地衣芽孢桿菌)發(fā)酵玉米蛋白粉的優(yōu)化條件。通過各菌種玉米蛋白粉水解馴化試驗、菌種混合比例及發(fā)酵條件(溫度、時間、含水量及初始p H)的正交試驗,以玉米蛋白粉多肽得率為指標(biāo),確定發(fā)酵玉米蛋白粉的最優(yōu)發(fā)酵條件。并在此條件下,測定發(fā)酵液中粗蛋白質(zhì)、總糖、還原糖、氨基酸含量以及多肽得率。結(jié)果顯示,正交試驗優(yōu)化的最佳接種比例為地衣芽孢桿菌4%、乳酸菌4%、酵母菌6%、黑曲霉6%,最佳發(fā)酵條件為發(fā)酵溫度35℃、發(fā)酵時間84 h、含水量45%、初始p H為6,在此條件下多肽得率為44.03%。在發(fā)酵過程中,粗蛋白質(zhì)含量和多肽得率呈上升趨勢,發(fā)酵終止時,粗蛋白質(zhì)含量提高了15.17%,多肽得率提高了205%,還原糖含量提高了850%,總必需氨基酸和總氨基酸含量分別增加了12.58%和12.37%。本試驗結(jié)果可為擴大玉米蛋白粉的應(yīng)用提供科學(xué)依據(jù)。
[Abstract]:The purpose of the experiment was to study the optimum conditions of fermentation of corn protein powder by compound bacteria (Aspergillus Niger, lactic acid bacteria, yeast and Bacillus licheniformis). Through the hydrolysis and acclimation test of corn protein powder of each strain, the orthogonal test of mixed proportion of bacteria and fermentation conditions (temperature, time, water content and initial pH), the polypeptide yield of corn protein meal was taken as the index. The optimum fermentation conditions of fermented corn protein powder were determined. Under these conditions, crude protein, total sugar, reducing sugar, amino acid content and polypeptide yield were determined. The results showed that the optimum inoculation ratio was Bacillus licheniformis 4, lactic acid bacteria 4, yeast 6, Aspergillus Niger 6. The optimum fermentation conditions were 35 鈩,
