不同脂肪酸對(duì)體外培養(yǎng)豬肌管細(xì)胞脂質(zhì)代謝的影響

【摘要】:本研究旨在探究日糧中不同飽和度脂肪酸棕櫚酸、油酸、亞油酸在體外培養(yǎng)的豬肌管細(xì)胞內(nèi)的代謝和沉積差異。體外分離培養(yǎng)豬骨骼肌衛(wèi)星細(xì)胞后,誘導(dǎo)成肌分化,分別用100μmol/L普通和[1-~(14)C]標(biāo)記棕櫚酸、油酸、亞油酸孵育豬肌管細(xì)胞24 h后,細(xì)胞甘油三酯測(cè)定、同位素示蹤法跟蹤3種脂肪酸的代謝過程,實(shí)時(shí)熒光定量和Western blot檢測(cè)脂質(zhì)代謝相關(guān)基因和蛋白的表達(dá)。結(jié)果表明:亞油酸組細(xì)胞中甘油三酯沉積量最高(P0.05);同位素示蹤法發(fā)現(xiàn)亞油酸在細(xì)胞中的氧化量和攝取量均最高(P0.05),且在細(xì)胞甘油三酯中的沉積量最高(P0.05);亞油酸能夠顯著提高肌管細(xì)胞脂肪酸攝取相關(guān)基因CD36、FABP4、FATP1,脂肪酸氧化相關(guān)基因CPT1、Cyt C、AMPKα2的表達(dá)(P0.05),以及線粒體功能相關(guān)蛋白NRF1表達(dá),并能夠顯著促進(jìn)甘油三酯合成相關(guān)基因DGAT2、FAS的表達(dá)(P0.05)。相比于飽和脂肪酸和單不飽和脂肪酸,多不飽和脂肪酸亞油酸能夠促進(jìn)豬肌管細(xì)胞脂肪沉積,并能促進(jìn)肌管細(xì)胞脂肪酸攝取、氧化、甘油三酯合成及促進(jìn)相關(guān)基因表達(dá)。
[Abstract]:The purpose of this study was to explore the metabolism and deposition differences of fatty acid palmitic acid, oleic acid and linoleic acid in pig myotube cells cultured in vitro. After isolation and culture of porcine skeletal muscle satellite cells, the myoplastic differentiation was induced. Palmitic acid, oleic acid and linoleic acid were incubated with 100 mol/L common and [1-~ (14) C], respectively. After h, the metabolic process of 3 fatty acids was traced by triglyceride and isotope tracer method. The expression of lipid metabolism related genes and proteins was detected by real-time fluorescence quantitative and Western blot. The results showed that the content of triglycerides in the cells of linoleic acid group was the highest (P0.05), and the amount of linoleic acid in the cells was detected by the same hormone tracing method. The amount was the highest (P0.05), and the amount of deposition in triglyceride was the highest (P0.05), and linoleic acid could significantly improve the expression of fatty acid uptake related genes CD36, FABP4, FATP1, the expression of CPT1, Cyt C, AMPK alpha 2 (P0.05), and the NRF1 expression of mitochondrial function related proteins, and can significantly promote triglycerides. Synthesis related gene DGAT2, FAS expression (P0.05). Compared to saturated fatty acids and monounsaturated fatty acids, polyunsaturated fatty acid linoleic acid can promote the fat deposition of porcine myotubular cells, and can promote the fatty acid uptake, oxidation, triglyceride synthesis and the promotion of phase related gene expression in myotubular cells.
