ERK信號(hào)轉(zhuǎn)導(dǎo)通路對(duì)家兔腦炎原蟲性腦肉芽腫形成的影響

【摘要】:為了探討ERK信號(hào)轉(zhuǎn)導(dǎo)通路對(duì)腦炎原蟲性腦肉芽腫形成的影響,應(yīng)用普通染色、特殊染色、免疫組織化學(xué)技術(shù)和Western-blot技術(shù)分別對(duì)腦炎原蟲性腦肉芽腫進(jìn)行觀察,分析腦炎原蟲性腦肉芽腫形成與信號(hào)轉(zhuǎn)導(dǎo)通路中神經(jīng)生長(zhǎng)因子(NGF)、酪氨酸蛋白激酶(TPK)、絲裂源活化蛋白激酶(MAPK)和細(xì)胞外信號(hào)調(diào)節(jié)激酶(ERK)等四種關(guān)鍵性蛋白的關(guān)系。結(jié)果顯示,所有患病家兔的腦組織中均有數(shù)量不等的腦肉芽腫和非化膿性腦炎變化;免疫組化染色顯示,NGF、TPK、MAPK和ERK的表達(dá)在肉芽腫中的星形膠質(zhì)細(xì)胞、血管內(nèi)皮細(xì)胞和成纖維細(xì)胞中明顯增高,呈強(qiáng)陽(yáng)性反應(yīng),染成深棕色;用Western-blot檢測(cè),患病家兔腦組織中的NGF、TPK、MAPK和ERK等信號(hào)轉(zhuǎn)導(dǎo)通路關(guān)鍵蛋白表豐度明顯高于健康家兔。以上結(jié)果表明,ERK信號(hào)轉(zhuǎn)導(dǎo)通路通過強(qiáng)大的級(jí)聯(lián)反應(yīng)參與了家兔腦炎原蟲性腦肉芽腫的形成,在腦肉芽腫形成過程中促進(jìn)星形膠質(zhì)細(xì)胞、血管內(nèi)皮細(xì)胞和成纖維細(xì)胞增殖。
[Abstract]:In order to investigate the effect of ERK signal transduction pathway on the formation of protozoan granuloma in encephalitis, general staining, special staining, immunohistochemical technique and Western-blot technique were used to observe the protozoan granuloma of the encephalitis, and the formation of brain granuloma and the nerve growth factor (NGF), casein in the signal transduction pathway of encephalitis protozoa were analyzed. The relationship between four key proteins such as acid protein kinase (TPK), mitogen activated protein kinase (MAPK) and extracellular signal regulated kinase (ERK). The results showed that all of the brain tissues of all the sick rabbits were different in brain granuloma and non suppurative encephalitis; immunohistochemical staining showed that the expression of NGF, TPK, MAPK and ERK in the granuloma The astrocytes, vascular endothelial cells and fibroblasts were significantly higher, and were strongly positive and dyed dark brown. The abundances of key proteins in the signal transduction pathways of NGF, TPK, MAPK and ERK in the brain tissues of the sick rabbits were significantly higher than those of healthy rabbits. The results showed that the ERK signal transduction pathway was stronger than that of the healthy rabbits. The cascade reaction participates in the formation of the protozoan granuloma of the rabbit encephalitis and promotes the proliferation of astrocytes, vascular endothelial cells and fibroblasts during the formation of brain granuloma.
