寧夏地區(qū)奶牛乳房炎病原菌分離鑒定及金葡菌毒力基因分析

【摘要】:本研究以寧夏銀川地區(qū)5個(gè)奶牛場(chǎng)、銀南地區(qū)7個(gè)奶牛場(chǎng)、銀北地區(qū)5個(gè)奶牛養(yǎng)殖場(chǎng)的94頭臨床型乳房炎奶牛、70頭隱性乳房炎奶牛為實(shí)驗(yàn)對(duì)象。臨床型乳房炎乳樣共分離鑒定出40種159株病原菌,其中葡萄球菌41株(25.79%)、鏈球菌6株(3.78%)、其他G+球菌20株(12%)、G-桿菌74株(46%)、酵母菌18株(11.32%)。隱性乳房炎乳樣共分離鑒定出33種共94株細(xì)菌,其中葡萄球菌28株(29.79%)、鏈球菌2株(2.13%)、其他G+球菌15株(15.96%)、G-桿菌45株(47.87%)、酵母菌4株(4.26%)。用15種抗細(xì)菌藥敏片對(duì)臨床型乳樣檢出率較高的12種細(xì)菌進(jìn)行藥敏試驗(yàn)。通過(guò)試驗(yàn)發(fā)現(xiàn),引起奶牛乳房炎的病原菌種類呈現(xiàn)多樣化、復(fù)雜化,與以往人們認(rèn)識(shí)的引起乳房炎的三種主要細(xì)菌為金黃色葡萄球菌、鏈球菌、大腸桿菌存有較大差距,且多數(shù)感染均為兩種或兩種以上細(xì)菌的混合感染。引起乳房炎的細(xì)菌種類常隨著地區(qū)的不同呈現(xiàn)不同的分布狀態(tài),即使同一地區(qū),甚至同一奶牛場(chǎng),致病菌的種類也會(huì)受操作規(guī)范、環(huán)境衛(wèi)生等方面的影響。對(duì)寧夏地區(qū)140株隱性乳房炎金黃色葡萄球菌的nuc、Hla、Hlb、fnbp A、fnbp B及9種腸毒素共14種毒力基因進(jìn)行檢測(cè)。實(shí)驗(yàn)結(jié)果顯示nuc、Hla、Hlb、fnbp A、fnbp B和SEI基因檢出率高,SEA、SEB、SEC、SED、SEH、SEG、SEJ檢出率較低或沒(méi)有檢出,可以提示nuc、Hla、Hlb、 fnbp A和SEI是寧夏地區(qū)隱性奶牛乳房炎金黃色葡萄球菌的主要毒力基因。
[Abstract]:In this study, 94 clinical mastitis cows with recessive mastitis were selected from 5 dairy farms in Yinchuan area, 7 dairy farms in south Yinnan area and 94 clinical mastitis cows in Yinbei area. A total of 40 species and 159 strains of pathogenic bacteria were isolated and identified from clinical mastitis, including 41 strains of Staphylococcus, 6 strains of Streptococcus, 20 strains of other G cocci, 74 strains of G- bacillus, and 18 strains of yeast, respectively. A total of 94 strains of 33 species were isolated from the milk samples of recessive mastitis, of which 28 strains were staphylococcus, 2 strains were streptococcus, 15 strains were isolated from 15 strains of G coccus, 45 strains were isolated from G- bacillus, and 4 strains were isolated from yeast. Fifteen antimicrobial drug sensitive tablets were used to test the drug sensitivity of 12 kinds of bacteria with high detection rate of clinical milk samples. It was found that the pathogenic bacteria causing mastitis of dairy cattle were diversified and complicated, which was different from the three main bacteria that caused mastitis were Staphylococcus aureus, Streptococcus aureus and Escherichia coli. And most of the infections were mixed infection of two or more kinds of bacteria. The species of bacteria causing mastitis often present different distribution with different regions. Even in the same area, even in the same dairy farm, the species of pathogenic bacteria will be affected by the operation standard, environmental hygiene and so on. A total of 14 virulence genes were detected in 140 strains of recessive mastitis Staphylococcus aureus in Ningxia. The results showed that the detection rate of Hla-Hla-Hlb-fnbp AfnBP B and SEI gene was high. The detection rate of SEDX SEDX SEHNGN SEJ was low or not, which suggested that nuclear Hlaa Hlb, fnbp A and SEI were the main virulence genes of recessive dairy cow mastitis Staphylococcus aureus in Ningxia area.
