鎘及α-硫辛酸對大鼠肝臟GJIC功能的影響

【摘要】:為探討鎘及α-硫辛酸(α-LA)對大鼠肝細(xì)胞縫隙通訊功能(GJIC)的影響,將30只100g左右SD雌性大鼠隨機(jī)均分為3組:對照組(自由飲水及超純水灌胃)、鎘組(50mg/L醋酸鎘自由飲水及超純水灌胃)、鎘與α-LA共同處理組(50mg/L醋酸鎘自由飲水及α-LA灌胃50mg/kg)。12周后,采用熒光染料標(biāo)記傳輸示蹤法(IL/DT)測定GJIC功能,免疫印跡檢測大鼠肝臟中Cx43、p-Cx43、Cx32蛋白表達(dá)變化,熒光定量PCR法檢測大鼠肝臟中Cx32、Cx26、Cx43mRNA表達(dá)量的變化。IL/DT結(jié)果顯示,大鼠長時間飲用醋酸鎘后,肝臟細(xì)胞間熒光黃兩側(cè)擴(kuò)散距離極顯著減少(P0.01),α-LA處理后極顯著提高(P0.01)。肝臟中Cx43、p-Cx43、Cx32蛋白表達(dá)量呈顯著或極顯著的降低,Cx43、Cx32、Cx26mRNA轉(zhuǎn)錄基因表達(dá)也呈顯著或極顯著的降低(P0.05或P0.01),α-LA處理后都有顯著或極顯著升高(P0.05或P0.01)。結(jié)果表明:長期飲用醋酸鎘可以抑制大鼠肝細(xì)胞GJIC,可能是連接蛋白或基因表達(dá)減少,α-LA能緩解鎘對GJIC的影響。
[Abstract]:To investigate the effects of cadmium and 偽 -lipoic acid (偽 -LAA) on gap communication function of rat hepatocytes, Thirty female Sprague-Dawley rats were randomly divided into three groups: control group (free drinking water and ultra-pure water), cadmium acetate free drinking water (50 mg / L) in cadmium group and ultra-pure water, cadmium acetate free drinking water and 偽 -LA treatment group (50 mg / L CD _ 2O _ 3 free drinking water and 偽 -LA) after 50mg/kg).12 week. The function of GJIC was detected by fluorescent dye transfer tracer method. Western blot was used to detect the expression of Cx43 p-Cx43 Cx32 protein in rat liver. The results of IL / DT showed that the expression of Cx32Cx26Cx43 mRNA in rat liver was detected by fluorescence quantitative PCR, and the results of IL / DT showed that after drinking cadmium acetate for a long time, the expression of Cx43 protein in rat liver was detected by fluorescence quantitative PCR. The diffusion distance between the two sides of the liver cells decreased significantly, and that of 偽 -LA increased significantly. In liver, the expression of Cx43, p-Cx43, Cx32 was significantly or extremely significantly decreased, and the expression of Cx43, Cx32, Cx26 mRNA was significantly or extremely significantly decreased by P0.01or P0.01. after 偽 -LA treatment, there was a significant or extremely significant increase in the mRNA expression of Cx43, Cx32 and Cx26, respectively (P0.05 or P0.01A) after treatment with 偽 -LA, there was a significant or extremely significant increase in the expression of Cx43, Cx32 and Cx26 mRNA, respectively. The results showed that drinking cadmium acetate for a long time could inhibit the expression of connexin or gene in rat hepatocytes, and 偽 -LA could alleviate the effect of cadmium on GJIC.
