【摘要】：建立動(dòng)物疫病的快速診斷,對(duì)于我國(guó)養(yǎng)殖業(yè)疫病的預(yù)防及控制、進(jìn)出口檢疫、動(dòng)物食品安全衛(wèi)生等方面具有重大意義。重組酶聚合酶擴(kuò)增技術(shù)(RPA)是近幾年出現(xiàn)的1種有望替代PCR的新的核酸擴(kuò)增技術(shù)。該技術(shù)主要依賴于3種酶:能結(jié)合單鏈寡核苷酸引物的重組酶、單鏈DNA結(jié)合蛋白和DNA聚合酶。反應(yīng)過程中不需要模板鏈的熱變性,可以在恒定的低溫條件下快速擴(kuò)增DNA或者RNA。具有特異性強(qiáng)、靈敏度高、快速、操作簡(jiǎn)便、適用于現(xiàn)場(chǎng)快速診斷等特點(diǎn),在動(dòng)物疾病早期診斷、實(shí)地檢測(cè)、進(jìn)出口快速檢疫等方面具有巨大的應(yīng)用前景和市場(chǎng)。本文對(duì)RPA技術(shù)進(jìn)行綜述,以期為相關(guān)研究提供參考。
[Abstract]:It is of great significance for the prevention and control of animal epidemic disease, import and export quarantine, animal food safety and health to establish a rapid diagnosis of animal epidemic disease in China. Recombinant enzyme polymerase amplification (rpa) is a new nucleic acid amplification technique which is expected to replace PCR in recent years. The technique mainly depends on three enzymes: recombinant enzyme that binds single strand oligonucleotide primer, single strand DNA binding protein and DNA polymerase. The thermal denaturation of the template chain is not required during the reaction, and the DNA or RNA can be rapidly expanded at constant low temperature. It has the characteristics of strong specificity, high sensitivity, rapid operation, and is suitable for field rapid diagnosis. It has a great application prospect and market in the early diagnosis of animal diseases, field detection, import and export rapid quarantine and so on. In this paper, RPA technology is reviewed in order to provide reference for related research.
