白羽王鴿GHR、MSTN基因多態(tài)性與生長(zhǎng)性能和屠宰性能的關(guān)聯(lián)性分析

【摘要】:生長(zhǎng)性狀和屠宰性狀是衡量動(dòng)物生產(chǎn)性能的兩個(gè)重要指標(biāo)。白羽王鴿作為我國(guó)優(yōu)異鴿種資源,深受廣大養(yǎng)殖戶的喜歡。本研究首先對(duì)白羽王鴿7個(gè)體尺指標(biāo)和7個(gè)屠宰指標(biāo)進(jìn)行測(cè)定,并對(duì)各指標(biāo)進(jìn)行了相關(guān)性分析,并選擇與生長(zhǎng)發(fā)育密切相關(guān)的GHR基因和MSTN基因?yàn)檠芯繉?duì)象,通過直接測(cè)序的方法掃描篩選其SNP位點(diǎn)(Single nucleotide polymorphism site,SNPs),分析白羽王鴿GHR、MSTN基因的單核苷酸多態(tài)性與生長(zhǎng)性狀之間的相關(guān)性,為白羽王鴿分子標(biāo)記輔助育種實(shí)踐提供理論依據(jù)。論文的主要研究結(jié)果和結(jié)論如下:(1)對(duì)白羽王鴿7個(gè)體尺指標(biāo)和7個(gè)屠宰指標(biāo)進(jìn)行測(cè)定,并對(duì)各指標(biāo)進(jìn)行了相關(guān)性分析。結(jié)果表明,相關(guān)性始終存在于體重、體尺與屠宰性狀之間:體重與除骨盆寬之外的其余6個(gè)體尺指標(biāo)(體斜長(zhǎng)、龍骨長(zhǎng)、胸深、胸寬、脛長(zhǎng)、脛圍)相關(guān)性均為正相關(guān),且相關(guān)性顯著或極顯著(P0.01)。對(duì)白羽王鴿體重與體斜長(zhǎng)、胸深、龍骨長(zhǎng)、骨盆寬,屠體重與體斜長(zhǎng)、胸深、龍骨長(zhǎng)、骨盆寬,全凈膛重與體斜長(zhǎng)、龍骨長(zhǎng),胸肌重與體斜長(zhǎng)、龍骨長(zhǎng)、骨盆寬、脛長(zhǎng),腿肌重與體斜長(zhǎng)、脛圍,腹肌重與體斜長(zhǎng)、胸寬分別建立了回歸模型,模型擬合度F檢驗(yàn)均達(dá)顯著或極顯著水平。(2)采用PCR-直接測(cè)序的方法對(duì)白羽王鴿GHR、MSTN的SNP位點(diǎn)進(jìn)行掃描,結(jié)果在GHR基因中發(fā)現(xiàn)6個(gè)突變位點(diǎn),分別為C264T、G410A、C992T、T1060C、G1160T、T1197A,在MSTN基因檢測(cè)到1個(gè)突變位點(diǎn),位于C114T處。由于堿基的突變,其中 C264T、C992T、T1060C、G1160T、T1197A 和 C114T 共 6 處突變位點(diǎn)改變了編碼的氛基酸種類。(3)運(yùn)用最小二乘法分析表明,GHR基因C264T位點(diǎn)的TT基因型個(gè)體的骨盆寬度極顯著低于CT和CC基因型個(gè)體(P0.01);GHR基因G410A位點(diǎn)的GG基因型個(gè)體的出生重量顯著高于GA基因型個(gè)體(P0.05);在脛圍性狀上,GHR基因C992T位點(diǎn)的TT基因型CC基因型CT基因型,且差異達(dá)到顯著水平(P0.05);GHR基因T1060C位點(diǎn)的不同基因型個(gè)體在各性狀上均未達(dá)到差異顯著性水平;GHR基因G1160T位點(diǎn)的GT基因型個(gè)體的脛長(zhǎng)長(zhǎng)度顯著性高于GG基因型個(gè)體(P0.05);GHR基因T1197A位點(diǎn)的TA基因型個(gè)體和TT基因型個(gè)體的骨盆寬度極顯著高于AA基因型個(gè)體(P0.01)。(4)運(yùn)用最小二乘法分析表明,MSTN基因C114T位點(diǎn)處CT基因型個(gè)體腹脂重顯著高于CC基因型個(gè)體(P0.05)。
[Abstract]:Growth traits and slaughter traits are two important indexes to measure the performance of animals. White feather pigeon as an excellent pigeon species resources, by the majority of farmers like. In this study, the seven individual size indexes and seven slaughtering indexes of white feather pigeon were measured, and the correlation of each index was analyzed. The GHR gene and MSTN gene, which were closely related to growth and development, were selected as the research objects. Single nucleotide polymorphism SNPs were screened by direct sequencing, and the correlation between single nucleotide polymorphisms of GHR-MSTN gene and growth traits was analyzed in order to provide theoretical basis for molecular marker-assisted breeding in white feather pigeon. The main results and conclusions of this paper are as follows: (1) the indexes of 7 individual scales and 7 slaughtering indexes of white feather pigeon were measured, and the correlation of each index was analyzed. The results showed that the correlation was always between body weight, body size and slaughter traits. There was a positive correlation between body weight and the other 6 individual measures except pelvis width (body oblique length, keel length, chest depth, chest width, tibial length, tibial circumference). The correlation was significant or extremely significant (P 0.01). The weight and body length of white feather pigeon, chest depth, keel length, pelvis width, butcher weight and body oblique length, chest depth, keel length, pelvic width, full bore weight and body oblique length, keel length, chest muscle weight and body oblique length, keel length, pelvis width, shank length, The regression models of leg muscle weight and body oblique length, tibial circumference, abdominal muscle weight and body oblique length, chest width were established respectively. The fit F test of the model was significant or extremely significant. The SNP site of GHRMSTN was scanned by PCR-direct sequencing. Results six mutation sites were found in the GHR gene, namely C264TG410AnC992T992 T1060CG1160TnT1197A, and one mutation locus was detected in the MSTN gene, which was located at C114T. The results showed that the pelvis width of TT genotype individuals with GHR gene C264T loci was significantly lower than that of individuals with CT and CC genotypes. The birth weight of GG genotype individuals at G410A locus was significantly higher than that of GA genotype individuals (P 0.05). The tibial length of GT genotype individuals with GHR gene G1160T locus was significantly higher than that of GG genotype individuals with different genotype T1060C loci. The tibial length of GT genotype individuals with GHR gene G1160T locus was significantly higher than that with GG genotype individuals. Because the pelvis width of TA genotype individuals and TT genotype individuals at T1197A locus was significantly higher than that of AA genotype individuals (P0.01U. 4) least square analysis showed that the abdominal fat weight of CT genotype individuals at C114T locus of MSTN gene was significantly higher than that of CC genotype individuals.
