番鴨呼腸孤病毒感染誘導(dǎo)細(xì)胞自噬的研究

【摘要】:本研究通過(guò)透射電鏡觀察自噬體的形態(tài)學(xué)結(jié)構(gòu),蛋白免疫印跡(Western blot)檢測(cè)細(xì)胞自噬相關(guān)蛋白LC3II和磷酸化mTOR(p-mTOR)表達(dá)量的動(dòng)態(tài)變化趨勢(shì),探索番鴨呼腸孤病毒(MDRV)感染對(duì)細(xì)胞自噬的影響。結(jié)果顯示,MDRV-YB株感染的DF-1細(xì)胞中可見(jiàn)典型的包裹胞漿成分的雙層膜結(jié)構(gòu);MDRV感染番鴨成纖維細(xì)胞(MDFs)和DF-1細(xì)胞后0~60h內(nèi),其LC3II表達(dá)量均呈現(xiàn)先升高后降低的趨勢(shì),且均在感染后36h達(dá)到最大值,其p-mTOR表達(dá)量均呈現(xiàn)先降低后回升的趨勢(shì),且分別在感染后36h和24h達(dá)到最低值;滅活MDRV組細(xì)胞未見(jiàn)LC3II表達(dá)。結(jié)果表明MDRV能夠誘導(dǎo)感染的MDFs和DF-1細(xì)胞持續(xù)較長(zhǎng)時(shí)間細(xì)胞自噬,而滅活的MDRV不能引起細(xì)胞自噬。
[Abstract]:In this study, the morphological structure of autophagy was observed by transmission electron microscope, and the expression of autophagy associated protein (LC3II) and phosphorylated mTOR-p-mTOR-mTor were detected by Western blot. The effect of muscovy duck reovirus (MDRV) infection on autophagy was investigated. The results showed that the expression of LC3II in DF-1 cells infected with MDRV-YB strain showed a tendency of increasing first and then decreasing within 60 h after infection with MDRV-YB strain and MDRV-infected muscovy duck fibroblasts (MDFs) and DF-1 cells. The p-mTOR expression reached the highest value at 36h after infection, and the expression of p-mTOR decreased first and then increased, and reached the lowest value at 36h and 24h after infection, and no LC3II expression was found in MDRV group. The results showed that MDRV could induce long-term autophagy of infected MDFs and DF-1 cells, while inactivated MDRV could not induce autophagy.
