犬鉤端螺旋體病PCR和熒光定量PCR診斷試劑盒的研制

【摘要】:鉤端螺旋體。ê(jiǎn)稱鉤體。且蚋腥揪哂兄虏⌒缘你^體所引起的一種人獸共患病。犬鉤端螺旋體病,是由常見(jiàn)對(duì)犬致病的犬型、黃疸出血型、塔拉索夫型等鉤體所引起。犬對(duì)該病具有易感性,犬感染本病后,臨床表現(xiàn)多樣,不易進(jìn)行診斷,因此需要實(shí)驗(yàn)室檢測(cè)對(duì)本病進(jìn)行輔助的鑒別診斷。常見(jiàn)實(shí)驗(yàn)室診斷方法有直接鏡檢,分離培養(yǎng),血清學(xué)檢測(cè),動(dòng)物接種等,但這些方法或不具備時(shí)效性,或缺乏準(zhǔn)確性。相較于這些方法,聚合酶鏈反應(yīng)(PCR)方法更省時(shí)省力,特異性和敏感性優(yōu)異,并且可用于鉤體病的早期診斷。 本實(shí)驗(yàn)依據(jù)僅致病性鉤端螺旋體含有的LigB基因序列的保守片段,并對(duì)常見(jiàn)對(duì)犬致病的鉤體株的相關(guān)序列進(jìn)行比對(duì),設(shè)計(jì)了數(shù)對(duì)特異性的引物和探針,目的片段大小為124bp。對(duì)探針和引物進(jìn)行篩選及反應(yīng)條件與體系的優(yōu)化,構(gòu)建了含目的基因的質(zhì)粒作為標(biāo)準(zhǔn)陽(yáng)性對(duì)照,,并對(duì)質(zhì)粒進(jìn)行測(cè)序驗(yàn)證。建立了犬鉤端螺旋體病PCR和熒光定量PCR診斷方法,并組裝成可實(shí)現(xiàn)對(duì)犬鉤體病進(jìn)行快速高效診斷的試劑盒。對(duì)試劑盒的靈敏度、特異性、重復(fù)性等參數(shù)進(jìn)行了檢驗(yàn),確定其保存條件和保存期,采集并檢測(cè)臨床犬血液和尿液樣本,與顯微凝集試驗(yàn)相比較,結(jié)果一致。 近年來(lái),寵物犬的數(shù)量不斷增加,各類工作犬的數(shù)量也有上升。犬易感鉤體病,影響犬自身及其主人的健康,該檢測(cè)方法可推廣應(yīng)用于犬鉤體病的早期檢測(cè),避免該病的進(jìn)一步發(fā)展和擴(kuò)散。 鉤端螺旋體的培養(yǎng)需要數(shù)周的時(shí)間,培養(yǎng)基的配制程序復(fù)雜,而診斷鉤體病的金標(biāo)準(zhǔn)方法MAT,需要活體鉤體才能進(jìn)行診斷,因而很難得到推廣和應(yīng)用,而PCR方法不需要進(jìn)行鉤體的培養(yǎng),并且相比于血清學(xué)方法,還可用于鉤體病的早期診斷。在該實(shí)驗(yàn)中,建立的普通PCR方法檢測(cè)鉤體濃度的靈敏度可達(dá)到101,熒光定量PCR方法的靈敏度可達(dá)100。
[Abstract]:The leptospirosis is a kind of zoonotic disease caused by the leptospirosis caused by infection . The canine leptospirosis is caused by the common dog type , jaundice , blood type and Tarasov type . The dog has a susceptibility to the disease . The dog has a variety of clinical manifestations and is not easy to diagnose . The common laboratory diagnosis method has direct microscopic examination , separation culture , serological detection , animal vaccination , etc . The method has the advantages of saving time and effort , specificity and sensitivity , and can be used for early diagnosis of leptospirosis .
The PCR and fluorescence quantitative PCR diagnosis of leptospirosis in dogs was established . The PCR and fluorescence quantitative PCR methods were used to test the sensitivity , specificity and repeatability of the kit . The blood and urine samples were collected and tested . The results were consistent with the microscopic agglutination test .
In recent years , the number of pet dogs has increased , and the number of various kinds of working dogs has also increased . The dog is susceptible to leptospirosis and affects the health of the dog itself and its owner . The detection method can be popularized and applied to the early detection of dog leptospirosis , so as to avoid further development and diffusion of the disease .
The culture of leptospirosis requires several weeks , the preparation procedure of the culture medium is complex , and the gold standard method MAT for diagnosing leptospirosis needs a living hook body for diagnosis , so it is difficult to be popularized and applied , and the PCR method does not need to carry out early diagnosis of leptospirosis compared with serological method .
