犬腫瘤診斷、治療及環(huán)氧合酶-2的表達(dá)

【摘要】:隨著我國(guó)犬的養(yǎng)殖數(shù)量日漸增多,隨之而來的犬病發(fā)生率也在上升,犬的腫瘤疾病就是其中之一,且有日漸增多的趨勢(shì),但尚未引起寵物飼養(yǎng)者的重視。為探討犬腫瘤臨床發(fā)病規(guī)律,在江浙滬地區(qū)部分寵物醫(yī)院門診收集了133例犬腫瘤自然發(fā)生病例,對(duì)其臨床癥狀、發(fā)病部位、品種、年齡、性別、飲食情況和轉(zhuǎn)移情況等進(jìn)行調(diào)查;用外科手術(shù)的方法摘除56例臨床自然發(fā)生的犬腫瘤,將病料固定、切片、染色后,通過顯微鏡觀察組織結(jié)構(gòu)和細(xì)胞形態(tài),并進(jìn)行腫瘤分類和病變類型;探討環(huán)氧合酶-2(COX-2)與犬腫瘤的發(fā)生、發(fā)展的關(guān)系,本研究收集術(shù)后的腫瘤組織46份和在浙江大學(xué)動(dòng)物醫(yī)院解剖的犬正常乳腺、皮膚、睪丸、肛周、子宮組織各6份,將上述組織包埋的蠟塊做成切片,應(yīng)用免疫組織化學(xué)S-P染色法檢測(cè)COX-2在46份犬腫瘤組織及30份正常組織中的表達(dá)情況。本試驗(yàn)得到以下結(jié)果:1調(diào)查門診收集的133例臨床自然發(fā)生的犬腫瘤病例,結(jié)果顯示:腫瘤發(fā)病早期一般不表現(xiàn)全身癥狀,惡性腫瘤發(fā)生轉(zhuǎn)移時(shí)會(huì)有惡病質(zhì)的表現(xiàn);乳腺和皮膚及軟組織是犬臨床高發(fā)的腫瘤部位,分別占全部患犬的42.11%和24.81%;京巴犬、雜交犬、貴賓犬、博美犬和西施犬是犬腫瘤高發(fā)的品種,分別占全部患犬的19.55%、14.29%、11.28%、8.27%和7.52%;患病犬年齡從1-15歲大小不等,平均年齡為9.39歲,6-10歲年齡段的患犬發(fā)病率最高;一些腫瘤類型與性別、是否絕育有關(guān);日常飲食與犬腫瘤的發(fā)生有關(guān),肉拌飯類、純?nèi)饧踊鹜饶c類、狗糧加肉類和純狗糧類,發(fā)病比例分別為39.85%、26.32%、21.05%和12.78%,其中純狗糧飼養(yǎng)的犬發(fā)病率明顯較低。2用外科手術(shù)的方法摘除56例臨床自然發(fā)生的犬腫瘤,結(jié)果顯示:外科手術(shù)是犬腫瘤最常用的治療方法,56例犬腫瘤中30例為惡性腫瘤,26例為良性腫瘤。根據(jù)腫瘤不同組織類型分類,29例乳腺腫瘤中惡性腫瘤21例,包括復(fù)合癌、導(dǎo)管乳頭狀癌、實(shí)質(zhì)型癌、未分化癌、黏液癌、纖維肉瘤、骨肉瘤和癌肉瘤;良性腫瘤8例,包括單純型腺瘤、復(fù)合型腺瘤、纖維腺瘤和乳腺增生。12例皮膚及軟組織腫瘤中惡性腫瘤3例,包括皮脂腺癌和脂肪肉瘤;良性腫瘤9例,包括基底細(xì)胞瘤、纖維瘤、血管周細(xì)胞瘤、皮脂腺瘤、皮脂腺上皮瘤和毛母細(xì)胞瘤。5例肛周腫瘤中惡性腫瘤3例為肛周腺癌;良性腫瘤2例為肛周腺瘤;6例陰道和子宮腫瘤中惡性腫瘤3例,包括纖維肉瘤和平滑肌肉瘤;良性腫瘤3例為平滑肌瘤。4例睪丸腫瘤都是良性腫瘤,包括支持細(xì)胞瘤和間質(zhì)細(xì)胞瘤。3 COX-2在犬腫瘤中的陽性表達(dá)主要位于腫瘤細(xì)胞的胞漿內(nèi)及胞核周圍,為棕黃色顆粒。46例犬腫瘤中30例COX-2呈強(qiáng)陽性表達(dá),5例呈陽性表達(dá),陽性表達(dá)率為76.1%(35/46);30份正常組織中COX-2的陽性表達(dá)率為20%(6/30)。COX-2在犬腫瘤和正常組織上的表達(dá)差異有統(tǒng)計(jì)學(xué)意義(P0.05)。46例進(jìn)行免疫組化染色的腫瘤中,19例良性腫瘤中15例COX-2呈強(qiáng)陽性表達(dá),陽性表達(dá)率為78.95%;27例惡性腫瘤中15例COX-2呈強(qiáng)陽性表達(dá),5例呈陽性表達(dá),陽性表達(dá)率為74.1%。COX-2在犬良、惡性腫瘤上的表達(dá)無統(tǒng)計(jì)學(xué)意義(P0.05)。提示COX-2與犬腫瘤的發(fā)生和發(fā)展有關(guān)系。
[Abstract]:With the increasing number of dog breeding in China, the incidence of canine disease is increasing, and the dog's tumor disease is one of them, and there is a growing trend, but it has not attracted the attention of the pet owners. In order to explore the clinical law of the dog's tumor, 133 cases of dog tumor were collected from the outpatient of the pet hospital in Jiangsu, Zhejiang and Shanghai. In case of cases, the clinical symptoms, location, variety, age, sex, diet and metastasis were investigated, and 56 cases of canine tumors that were naturally occurring were removed by surgical procedure, and the tissue structure and cell morphology were observed by microscope, and the tumor classification and pathological type were observed by microscope. To investigate the relationship between cyclooxygenase -2 (COX-2) and the occurrence and development of canine tumor. This study collected 46 tumor tissues after operation and 6 tissues of normal breast, skin, testis, perianal and uterus dissected at the animal hospital of Zhejiang University. The wax blocks embedded in the above tissue were cut into slices, and COX-2 was detected by immunohistochemical S-P staining method in 46. The following results were obtained in this test: 1 the results of 133 cases of naturally occurring canine tumor cases collected in the outpatient clinic were investigated. The results showed that the early stage of the tumor usually did not show systemic symptoms, and the malignant tumor appeared when the malignant tumor was transferred; the breast and skin and soft tissue were the dog's face. The high incidence of tumours in the bed accounted for 42.11% and 24.81% of the total dogs; Beijing, hybrid, poodle, BBO and Xishi were the high incidence of canine tumors, which accounted for 19.55%, 14.29%, 11.28%, 8.27% and 7.52% of all dogs. The age of the sick dog ranged from 1-15 years to 9.39 and 6-10 years old. The incidence of some tumor types was related to sex and sterilization; daily diet was associated with the occurrence of canine tumors, meat mixing, pure meat and ham sausage, dog food plus meat and pure dog food, respectively, 39.85%, 26.32%, 21.05% and 12.78%, respectively. The incidence of pure dog food in dogs was significantly lower than that of 56 cases of.2 surgery. The results showed that surgery was the most common treatment for canine tumors, 30 of 56 cases of canine tumors were malignant and 26 were benign. According to the classification of different tissue types, 21 cases of malignant tumor in 29 breast tumors, including compound carcinoma, ductal papillary carcinoma, parenchyma carcinoma, undifferentiated carcinoma, mucus cancer, fiber 8 cases of benign tumors, including simple adenoma, compound adenoma, fibroadenoma, and mammary gland hyperplasia in 3 cases of.12 and soft tissue tumors, including sebaceous adenocarcinoma and liposarcoma, and 9 cases of benign tumor, including basal cytoma, fibroma, pericytoma, sebaceous adenoma, and sebaceous epithelioma. And 3 cases of malignant tumor of perianal tumor in.5 cases, 2 cases of benign tumor were perianal adenoma, 6 cases of malignant tumor in vaginal and uterine tumor, 3 cases of malignant tumor, including fibrosarcoma and smooth muscle sarcoma, 3 cases of benign tumor and.4 case of leiomyoma, benign tumor, including support cell tumor and interstitial cell tumor.3 COX-2 The positive expression in the canine tumor was mainly located in the cytoplasm and the nucleus of the tumor cells. 30 cases of COX-2 were strongly positive in.46 cases of brown yellow granules. The positive expression was 76.1% (35/46), and the positive expression rate of COX-2 in 30 normal tissues was 20% (6/30).COX-2 in canine tumor and normal tissue. The difference was statistically significant (P0.05) in the tumor of.46 immunohistochemical staining, 15 cases of benign tumors were strongly positive in 19 cases of benign tumor, and the positive expression rate was 78.95%. 15 cases of COX-2 were strongly positive in 27 cases of malignant tumor and 5 cases were positive, the positive expression rate was 74.1%.COX-2 in canine, and there was no statistical significance in the malignant tumor. P0.05), suggesting that COX-2 is related to the occurrence and development of tumor in dogs.
