低劑量奶牛性控冷凍精液生產(chǎn)新技術(shù)產(chǎn)品試制與示范應(yīng)用

【摘要】:流式細胞儀分離精子的人工授精是目前控制哺乳動物出生后代性別最為可靠的繁育生物技術(shù)。但受專利和生產(chǎn)成本的限制,以奶牛為主的性控冷凍精液的高成本制約其廣泛推廣應(yīng)用,因此提高奶牛性控冷凍精液精液的生產(chǎn)效率是當(dāng)前降低產(chǎn)品成本的有效途徑。本研究通過研制一種混合異種動物精液的新型奶牛性控冷凍精液生產(chǎn)技術(shù),達到提高奶牛性控冷凍精液精生產(chǎn)效率、降低其生產(chǎn)成本并且可以進行產(chǎn)業(yè)化推廣應(yīng)用的目的。本實驗對比了3種劑型混合異種動物精液的奶;旌闲钥乩鋬鼍簝龊蠡盍、存活時間、頂體完整率檢測結(jié)果及成年母牛人工授精效果。首先以低劑量(100萬/0.25ml)奶牛分離X精子為性控冷凍精液產(chǎn)品目的精子,在分離X精液中添加不同濃度山羊精液,制備3個劑型的混合奶牛性控冷凍精液X1型、X2型、X3型(X1型含山羊精子300萬個/0.25ml,X2型含山羊精子600萬個/0.25ml,X3型含山羊精子900萬個/0.25ml)。然后對3個劑型凍精活力、存活時間、頂體完整率進行評定,來探討混合奶牛冷凍精液中不同濃度山羊精液對奶牛性控冷凍精液品質(zhì)以及人工授精的受胎率影響。實驗結(jié)果表明:X1型奶牛性控冷凍精液存活時間顯著高于X2型和X3型產(chǎn)品(8 vs 6 vs 5) P0.05; X1型奶牛性控冷凍精液活力較X2型和X3型有所提高(55% vs 52% vs 48%),但相互之間差異不顯著P0.05;X1型奶牛性控冷凍精液與X2型、X3型凍精的頂體完整率差異不顯著(54% vs52.7% vs 51.9%) P0.05。但是,X1型奶牛性控冷凍精液進行人工授精90天后的受胎率顯著高于X2型和X3凍精產(chǎn)品(51.9%vs33.6%vs29.9%) P0.05。根據(jù)上述實驗結(jié)果提出本研究結(jié)論:濃度為300萬/0.25ml的山羊精液混合100萬/0.25ml的分離奶牛X精液制備的混合奶牛性控冷凍精液新技術(shù)產(chǎn)品,使生產(chǎn)效率提高2倍相對生產(chǎn)成本降低50%,凍精質(zhì)量指標及人工授精的受胎率結(jié)果均達到常規(guī)性控冷凍精液的水平,證明該混合奶牛性控冷凍精液可以產(chǎn)業(yè)化生產(chǎn)并且進行產(chǎn)業(yè)化推廣應(yīng)用。
[Abstract]:The artificial insemination of separated sperm by flow cytometry is the most reliable reproductive biological technology to control the sex of the offspring of mammals. However, limited by the cost of the patent and production, the high cost of the frozen semen mainly controlled by the cow restricts its extensive application. Therefore, the efficiency of improving the production efficiency of the semen of milk cow's frozen semen is to be improved. The effective way to reduce the cost of the product is to improve the production technology of a new type of dairy cow sex controlled frozen semen, which is mixed with the semen of different species of animal, to improve the efficiency of the sperm production, reduce the cost of production and promote the popularization and application of the 3 kinds of mixture. The vitality, survival time, acrosome integrity test results and artificial insemination effect of adult cows were detected by mixed control frozen semen of physical semen. First, X sperm was isolated from dairy cows with low dose (1 million /0.25ml) as the target sperm of sex controlled frozen semen, adding different concentration of goat semen in the separation of X semen to prepare 3 dosage forms of mixed milk. Bovine sex controlled cryopreservation semen X1, X2, X3 (X1 type goat spermatozoa 3 million /0.25ml, X2 type goat spermatozoon 6 million / 0.25ml, X3 type goat spermatozoon 9 million /0.25ml), and then evaluate the vitality, survival time and acrosome integrity of 3 dosage forms of frozen semen to explore the performance of goat semen of different concentration in the frozen semen of mixed cows. The effects of the quality of frozen semen and the pregnancy rate of artificial insemination were controlled. The experimental results showed that the survival time of sex controlled frozen semen of X1 dairy cows was significantly higher than that of X2 and X3 type products (8 vs 6 vs 5) P0.05, and the activity of sex controlled cryopreservation semen in X1 dairy cows was higher than that of X2 and X3 (55% vs 52% vs 48%), but there was no significant difference between each other. There was no significant difference in the complete rate of the acrosome between the cow sex controlled cryopreservation semen and the X2 type and the X3 type freeze-cream (54% vs52.7% vs 51.9%) P0.05., but the abortion rate of the X1 type dairy cattle controlled frozen semen for 90 days was significantly higher than that of the X2 type and X3 cryopreservation product (51.9%vs33.6%vs29.9%) P0.05. based on the results mentioned above: the concentration was 3 million /0.25ml goat semen mixed with 1 million /0.25ml, a new technology product of mixed dairy cow sex controlled frozen semen prepared from dairy cow X semen, the production efficiency increased by 2 times the relative production cost by 50%, the quality index of the frozen semen and the artificial insemination rate reached the level of the conventional controlled cryopreservation semen, which proved that the mixed cow sex control was controlled. Frozen semen can be industrialized and popularized.
