【摘要】：為了解新疆尉犁縣庫蠓體內(nèi)攜帶病毒情況,從該縣采集18 000只庫蠓,50只為一組研磨后分別接種于BHK-21、C6/36、Vero細胞,盲傳5代,一組樣品使BHK-21出現(xiàn)CPE,先用5-氟尿核苷藥敏試驗鑒定為RNA病毒,之后用蟲媒RNA病毒引物擴增,鑒定為甲病毒,疑似蓋塔病毒,最后用中和試驗、免疫電鏡觀察和蓋塔病毒C基因特異性引物PCR等方法鑒定分離毒株為蓋塔病毒。本研究首次從新疆庫蠓體內(nèi)分離到蓋塔病毒,該毒株與2005年日本分離株相似性高達98%,推測可能是由于引進日本帶病種公畜或精液有關(guān),提示在進出口畜牧貿(mào)易活動中,應(yīng)加強蓋塔病毒的檢疫工作。
[Abstract]:In order to understand the virus transmission in Culicoides in Yuli County, Xinjiang, 18 000 Culicoides were collected from this county and inoculated into BHK-21 C6 / 36 Vero cells for 5 passages. A group of samples made BHK-21 appear RNA virus, first identified as RNA virus by 5-fluorouracil drug sensitivity test, then amplified with insect-mediated RNA virus primer, identified as A virus, suspected Gaeta virus, and finally used neutralization test. The isolated strain was identified as Gaeta virus by immunoelectron microscopy and specific primer PCR. In this study, Gaeta virus was first isolated from Culicoides kulicoides in Xinjiang. The similarity between the virus strain and the Japanese isolate in 2005 is as high as 98 kori, which suggests that the virus may be related to the introduction of Japanese diseased species, male animals or semen, suggesting that it is involved in the import and export of livestock trade. The quarantine of Gaeta virus should be strengthened.
