nisin及大黃復方對子宮內膜炎大鼠血清中MMP-2和MMP-9活性的影響

【摘要】:為研究乳酸鏈球菌素(nisin)及大黃復方對患大鼠子宮內膜炎血清中基質金屬蛋白酶-2、基質金屬蛋白酶-9(MMP-2、MMP-9)的作用,構建大鼠子宮內膜炎模型,將280只SD大鼠隨機分為nisin組、低、中、高劑量大黃復方組、陽性對照組、生理鹽水組以及模型組,建子宮內膜炎模型前0h和建模后24、48、72h,用ELISA試劑盒檢測各試驗組中基質金屬蛋白酶(MMPs)和基質金屬蛋白抑制物(TIMPs)的變化情況來確定治療效果。結果表明:1)在0~72h之間,模型組中,MMP-9和TIMP-1的質量濃度顯著低于生理鹽水組(P0.05),且MMP-2和TIMP-2的質量濃度逐漸降低;2)nisin和高劑量大黃復方組中MMP-9、TIMP-1、MMP-2和TIMP-2的質量濃度均顯著高于生理鹽水組(P0.05),其中以nisin組和高劑量大黃復方組效果最佳。由此可見,nisin和高劑量大黃復方能顯著提高子宮內膜炎大鼠血清中TIMP-1和TIMP-2的表達,抑制MMP-9和MMP-2的活性而發(fā)揮調控子宮內生理機能的作用,表明nisin、中、高劑量大黃復方對MMP-9、TIMP-1和MMP-2的質量濃度具有正調控作用。
[Abstract]:In order to study the effects of nisin) and rhubarb compound on matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-2MMP-9) in serum of endometritis rats, 280 SD rats were randomly divided into nisin group, low group and middle group. High dose rhubarb compound group, positive control group, normal saline group and model group, The changes of matrix metalloproteinase (MMPs) and matrix metalloproteinase inhibitor (TIMPs) were detected by ELISA kit in order to determine the therapeutic effect. The results show that 1) is between 0 and 72 hours. The mass concentrations of MMP-9 and TIMP-1 in the model group were significantly lower than those in the saline group (P 0.05), and the mass concentrations of MMP-2 and TIMP-2 decreased gradually, and the mass concentrations of MMP-9 TIMP-1MMP-2 and TIMP-2 in the high dose rhubarb compound group were significantly higher than those in the saline group, especially in the nisin group. And high dose rhubarb compound group is the best. It can be seen that nisin and high dose rhubarb compound can significantly increase the expression of TIMP-1 and TIMP-2 in the serum of endometritis rats, inhibit the activity of MMP-9 and MMP-2, and play a role in regulating the physiological function of uterus. High dose rhubarb compound has positive effect on the mass concentration of MMP-9 and TIMP-1 and MMP-2.
