H10亞型和N8亞型禽流感病毒雙重RT-PCR檢測方法的建立

【摘要】:根據(jù)Gen Bank中H10亞型和N8亞型禽流感病毒(AIV)的HA和NA基因保守序列,分別設(shè)計篩選出2對特異性引物,用于H10亞型和N8亞型AIV的檢測,優(yōu)化引物之間的濃度,建立了同時檢測H10亞型和N8亞型AIV的雙重RT-PCR檢測方法。該方法對H10N8亞型AIV可特異性擴(kuò)增出267 bp(H10亞型)和464 bp(N8亞型)目的條帶,對H10Ny(y≠8)亞型AIV僅擴(kuò)增出267 bp目的條帶,對HXN8(x≠10)亞型AIV僅擴(kuò)增出464 bp目的條帶,對其他亞型AIV和常見禽病病原體均未擴(kuò)增出任何條帶。該方法對H10亞型和N8亞型AIV檢測下限均為104拷貝數(shù)/μL。本研究建立的H10亞型和N8亞型AIV雙重RT-PCR特異性強(qiáng)、靈敏度高,可同時快速檢測H10亞型和N8亞型AIV,為其感染的快速鑒別診斷提供一種簡便、快速和有效的方法。
[Abstract]:According to the conserved sequences of HA and na genes of H10 subtype and N8 subtype avian influenza virus in Gen Bank, two pairs of specific primers were designed and screened for the detection of H10 subtype and N8 subtype AIV, and the concentration between primers was optimized.A dual RT-PCR method for simultaneous detection of H 10 subtype and N 8 subtype AIV was established.The target bands of H10N8 subtype AIV and 464 bp(N8 subtype were amplified by this method, only 267bp for H10Ny(y 鈮,
