【摘要】：【目的】沙門(mén)氏菌(Salmonella)是重要的人畜共患傳染病原菌,其感染是引起世界性胃腸疾病的主要因素,全球每年大概有2 100萬(wàn)傷寒病例,給世界公共衛(wèi)生帶來(lái)嚴(yán)重威脅。目前抗生素濫用問(wèn)題嚴(yán)峻,急需尋找一種抗生素的替代品。抗菌肽JH-3是本實(shí)驗(yàn)室分離并人工改造后具有廣譜殺菌活性的小肽,以沙門(mén)氏菌標(biāo)準(zhǔn)菌株CVCC541為研究對(duì)象,在小鼠模型上評(píng)價(jià)抗菌肽JH-3治療沙門(mén)氏菌感染的效果�！痉椒ā吭贑VCC541感染BALB/c小鼠前3 d連續(xù)腹腔注射抗菌肽JH-3或環(huán)丙沙星(B3d,共計(jì)40 mg/kg)和感染后3 d連續(xù)注射JH-3或環(huán)丙沙星(P3d,共計(jì)40 mg/kg)進(jìn)行治療。【結(jié)果】發(fā)現(xiàn)環(huán)丙沙星預(yù)防組效果最佳,抗菌肽JH-3的預(yù)防組(B3d)效果較好,可顯著保護(hù)小鼠免受致死劑量CVCC541的攻擊,小鼠存活率高達(dá)100%,臨床癥狀評(píng)分、血液和臟器荷菌數(shù)降低,小腸段病理變化減輕,效果與環(huán)丙沙星治療組相當(dāng);而感染3 d后JH-3治療效果較差,臨床癥狀評(píng)分、臟器荷菌量以及小腸病理變化均顯著高于3 d前預(yù)防組,但3 d后治療組小鼠存活率為70%,仍明顯高于單獨(dú)感染組。【結(jié)論】系統(tǒng)評(píng)價(jià)了抗菌肽JH-3不同給藥時(shí)間對(duì)沙門(mén)氏菌感染的治療效果,預(yù)防性給藥方式的抗菌作用最佳,與環(huán)丙沙星治療效果相當(dāng),為新型抗菌藥物的研究提供參考。
[Abstract]:[objective] Salmonella is an important zoonotic pathogen, and its infection is the main cause of gastrointestinal diseases worldwide. There are about 2 1 million cases of typhoid fever in the world every year, which pose a serious threat to public health in the world.At present, the problem of antibiotic abuse is serious, and it is urgent to find a substitute for antibiotics.Antimicrobial peptide JH-3 is a small peptide with broad-spectrum bactericidal activity after isolation and artificial modification in our laboratory. CVCC541, a standard strain of Salmonella, was used as the research object.To evaluate the efficacy of antimicrobial peptide JH-3 in the treatment of Salmonella infection in mice model. [methods] JH-3 or ciprofloxacin were injected intraperitoneally 3 days before CVCC541 infection in BALB/c mice for a total of 40 mg / kg) and JH-3 was injected continuously 3 days after infection.Ciprofloxacin P3d, 40 mg / kg, was treated. [results] it was found that ciprofloxacin prophylaxis group had the best effect.The prophylactic effect of antimicrobial peptide JH-3 was better than that of control group (B3d), which could significantly protect mice from lethal dose of CVCC541. The survival rate of mice was as high as 100. The clinical symptom score, the number of blood and organs were decreased, and the pathological changes of intestinal segment were alleviated.The effect was similar to that of ciprofloxacin group, but the effect of JH-3 was worse after 3 days of infection, and the clinical symptom score, organ load and intestinal pathological changes were significantly higher than those in the prevention group before 3 days.But after 3 days, the survival rate of mice in the treatment group was 70, which was still significantly higher than that in the single infection group. [conclusion] the therapeutic effect of antimicrobial peptide JH-3 on Salmonella infection was evaluated systematically, and the antibacterial effect of prophylactic administration was the best.The efficacy of ciprofloxacin is comparable to that of ciprofloxacin, which provides a reference for the study of new antimicrobial agents.
