單色綠光對雞胚褪黑激素合成的影響

【摘要】:本研究旨在探討單色綠光對雞胚松果體發(fā)育以及褪黑激素合成的影響,為研究單色綠光影響雞胚發(fā)育提供依據(jù),選用120枚質(zhì)量為(65±3)g的受精雞胚,隨機分為兩組,分別在綠光(波長560nm,光強15lx)和黑暗條件下孵化,每組設(shè)4個重復(fù),每個重復(fù)5枚雞胚,分別在E(胚齡)15、E18和E21取材,探索雞胚孵化過程中單色綠光對松果體的發(fā)育以及褪黑激素合成的影響。結(jié)果表明:(1)孵化期間給予綠光照射,雞胚質(zhì)量增加11.19%~21.41%(P0.05);(2)單色綠光促進松果體小葉發(fā)育;(3)褪黑激素合成關(guān)鍵酶——乙;D(zhuǎn)移酶(AANAT)轉(zhuǎn)錄水平的表達隨著胚齡的增加而升高,其中黑暗組E21時AANAT mRNA表達較E15和E18時顯著高29.84%~47.06%(P0.05),但是E15與E18間差異不顯著(P0.05);而綠光組中,E18 mRNA水平較E15 AANAT mRNA的表達顯著高17.76%(P0.05),E21時較E15和E18時松果體中AANAT mRNA表達水平顯著高26.37%~48.81%(P0.05);并且單色綠光組高于黑暗組12.85%~15.95%(E18~E21,P0.05);(4)血漿褪黑激素含量隨著胚齡的增加而升高,黑暗組E21褪黑激素含量為E15和E18的2.6和1.9倍,綠光組中E21為E15和E18的2.8和1.8倍;在E21和E18,綠光組雞胚外周血褪黑激素水平高于黑暗組的13.13%和18.20%(P0.05),并且與外周血中生長激素水平極顯著正相關(guān)(P0.001)。由此可知,在雞胚孵化期間給予單色綠光刺激可促進松果體發(fā)育以及褪黑激素的合成與釋放,進而促進生長激素水平,最終促進雞胚發(fā)育。
[Abstract]:The purpose of this study was to investigate the effects of monochromatic green light on the pineal development and melatonin synthesis of chicken embryos. In order to study the effect of monochromatic green light on the development of chicken embryos, 120 fertilized chicken embryos with a mass of 65 鹵3 g were randomly divided into two groups.Hatched under the conditions of green light (wavelength 560 nm, light intensity 15lx) and dark conditions, each group was divided into 4 replicates, each replicating 5 chicken embryos.To explore the effect of monochromatic green light on pineal development and melatonin synthesis during hatching of chicken embryo.The results showed that the weight of chicken embryo increased by 11.19% and 21.41% during hatching (P0.05) green light promoted the development of pineal lobule.
