AFB1誘導(dǎo)肉雞亞慢性肝損傷模型的建立及姜黃素的干預(yù)效果

【摘要】:肝臟是機(jī)體內(nèi)重要的解毒器官,攝入的毒物吸收后進(jìn)入肝臟通過(guò)生物轉(zhuǎn)化起到解毒或活化的作用。所以肝臟易受內(nèi)外源性毒物的侵害造成肝損傷。黃曲霉毒素B1(AFB1)是黃曲霉毒素(AF)中的一種,具有極強(qiáng)的肝毒性和致癌性。肉雞對(duì)AFB1易感。深入研究AFB1致肉雞肝損傷的發(fā)病機(jī)理,積極開發(fā)經(jīng)濟(jì)價(jià)值高、活性強(qiáng)、療效好的保肝藥物是我們重要的科研目標(biāo)。姜黃素是姜黃的主要活性成分,是天然的食品添加劑、抗氧化劑和抗炎藥物。本試驗(yàn)選擇國(guó)內(nèi)廣泛飼養(yǎng)的AA肉雞作為試驗(yàn)動(dòng)物,以臨床癥狀、肝體指數(shù)、血清轉(zhuǎn)氨酶(ALT、AST)、肝組織氧化應(yīng)激指標(biāo)(GSH-Px、SOD、MDA)及病理組織學(xué)作為評(píng)價(jià)指標(biāo),在篩選AFB1誘導(dǎo)AA肉雞亞慢性肝損傷的最佳濃度和時(shí)間的基礎(chǔ)上,建立AFB1誘導(dǎo)的AA肉雞亞慢性肝損傷模型。通過(guò)染毒的同時(shí)干預(yù)不同劑量的姜黃素,分析姜黃素對(duì)AFB1誘導(dǎo)的AA肉雞亞慢性肝損傷的干預(yù)效果。本試驗(yàn)結(jié)果對(duì)防治AA肉雞AFB1亞慢性中毒,研究AFB1致毒機(jī)制以及姜黃素在獸醫(yī)臨床上的應(yīng)用提供理論依據(jù)。研究結(jié)果表明:1、AFB1誘導(dǎo)AA肉雞亞慢性肝損傷模型的建立采用高(5 mg·kg~(-1))、中(3 mg·kg~(-1))、低(1 mg·kg~(-1))劑量AFB1添加入基礎(chǔ)飼料中,連續(xù)染毒AFB1 14 d、21 d、28 d。結(jié)果顯示,連續(xù)染毒28d,AFB1高劑量組雞肝體指數(shù)顯著高于空白對(duì)照組(P0.05);血清轉(zhuǎn)氨酶活性極顯著高于空白對(duì)照組(P0.01);肝組織內(nèi)抗氧化酶活性極顯著低于空白對(duì)照組(P0.01),氧化產(chǎn)物含量極顯著高于空白對(duì)照組(P0.01)。病理組織學(xué)結(jié)果顯示,AFB1高劑量組肝臟結(jié)構(gòu)紊亂,肝臟局部纖維化,血管內(nèi)淤血,肝索結(jié)構(gòu)模糊,肝細(xì)胞空泡變性,淋巴細(xì)胞浸潤(rùn)。中、高劑量AFB1能夠顯著降低AA肉雞平均日增重。提示連續(xù)28 d飼喂含有5 mg·kg~(-1) AFB1的飼料可以成功建立AA肉雞亞慢性肝損傷模型,AA肉雞出現(xiàn)明顯AFB1中毒癥狀、體重降低。2、姜黃素對(duì)AFB1致AA肉雞亞慢性肝損傷的干預(yù)效果本試驗(yàn)以臨床癥狀、肝體指數(shù)、血清轉(zhuǎn)氨酶活性、肝臟氧化應(yīng)激指標(biāo)及病理組織學(xué)結(jié)果為檢測(cè)項(xiàng)目,對(duì)飼料中添加不同劑量姜黃素(150 mg·kg~(-1)、300 mg·kg~(-1)和450 mg·kg~(-1))對(duì)AFB1誘導(dǎo)的AA肉雞亞慢性肝損傷的干預(yù)效果進(jìn)行評(píng)價(jià)。連續(xù)染毒及干預(yù)28 d,與模型組相比,姜黃素干預(yù)后,AA肉雞AFB1中毒癥狀減少,平均日增重增加,肝損傷明顯減輕,肝損傷緩解程度與姜黃素劑量呈正相關(guān)。說(shuō)明姜黃素連續(xù)干預(yù)的情況下,姜黃素對(duì)AFB1誘導(dǎo)的AA肉雞亞慢性肝損傷具有預(yù)防性的保護(hù)作用。停止染毒和姜黃素干預(yù)后,試驗(yàn)35 d,AFB1模型組肝臟仍存在明顯的肝損傷,高劑量姜黃素仍對(duì)AFB1誘導(dǎo)的AA肉雞亞慢性肝損傷存在持續(xù)的保護(hù)作用。以上結(jié)果表明,以5 mg·kg~(-1)劑量將AFB1拌入料中,連續(xù)飼喂染毒飼料28 d可成功建立AFB1誘導(dǎo)AA肉雞亞慢性肝損傷模型;姜黃素能夠有效拮抗AFB1所致AA肉雞亞慢性肝損傷,起到保護(hù)肝臟的作用,尤其是高劑量姜黃素的保護(hù)作用較為持久,可以有效緩解AFB1的毒性。建議混飼的姜黃素干預(yù)劑量為450 mg·kg~(-1)。
[Abstract]:The liver is an important detoxification organ in the body, intake of poison absorbed into the liver through biotransformation to detoxification or activation. So the liver is susceptible to exogenous toxins damage caused liver injury. Aflatoxin B1 (AFB1) is yellow aspergillus toxin (AF) in one, with strong liver toxicity and carcinogenicity. Chickens are susceptible to AFB1. The pathogenesis of liver injury caused by meat AFB1 is studied deeply, and actively develop high economic value, high activity, good effect of hepatoprotective drugs is our important research goal. Curcumin is the main active component of turmeric, is a natural food additive, antioxidant and anti-inflammatory drugs. This experiment a wide choice of domestic AA broiler breeding as test animal, with clinical symptoms, liver index, serum transaminase (ALT, AST), the index of oxidative stress in liver tissue (GSH-Px, SOD, MDA) and histopathology as the evaluation index, the selection of AFB 1 AA broilers induced by sub chronic liver injury of optimal concentration and time, the establishment of AA broiler sub chronic liver injury model induced by AFB1. Different doses of curcumin through exposure and intervention, analysis of curcumin intervention effect on the injury of AA broiler sub chronic liver induced by AFB1. The test results on the prevention and treatment of AA broilers AFB1 chronic poisoning and provide a theoretical basis for the application of AFB1 and the toxic mechanism of curcumin in veterinary clinic. The results showed that: 1, the establishment of the AA chicken sub chronic liver injury model was induced by high AFB1 (5 mg - kg~ (-1), (3) in mg kg~ (-1), low (1 mg) kg~ (-1)) dose of AFB1 added to the basal diet, and continuous exposure to AFB1 14 d, 21 d, 28 d. results showed that continuous exposure to 28d, AFB1 high dose group liver index were significantly higher than the control group (P0.05); serum transaminase activity was significantly higher than control group (P0.01); liver tissue Anti oxidase activity was significantly lower than control group (P0.01), the oxidation product content was significantly higher than the control group (P0.01). The pathological results showed that high dose of AFB1 group of liver structure disorder, liver fibrosis, vascular congestion, livercable structure fuzzy, liver cell degeneration, lymphocyte infiltration, high doses of AFB1 AA can significantly reduce the average daily gain of broiler. The results suggest that the 28 d feed containing 5 mg (-1) - kg~ AFB1 feed can be successfully established AA broiler sub chronic liver injury model, AA broilers appeared AFB1 poisoning symptoms, body weight decreased.2, intervention effect of curcumin on AFB1 induced sub chronic liver injury of AA broilers in the test the clinical symptoms, liver index, serum transaminase activity and oxidative stress in the liver index and histopathological results for the detection of the project, adding different doses of dietary curcumin (150 mg - kg~ (-1), 300 mg - kg~ (-1) and 4 50 mg - kg~ (-1)) to evaluate the intervention effect on the injury of AA broiler sub chronic liver induced by AFB1. Continuous exposure and 28 d intervention, compared with the model group, curcumin intervention, AA broiler AFB1 poisoning symptoms decreased, average daily gain, liver damage significantly reduced liver injury relief degree and curcumin the dose was positively correlated. Curcumin intervention, curcumin has a protective effect on the injury of AA broiler preventive sub AFB1 induced chronic liver. Stop exposure and curcumin intervention, 35 d test, AFB1 model group liver still exist significant liver injury, high dose of curcumin has protective effect on the injury of AA continuous broiler sub chronic liver induced by AFB1. The above results showed that 5 mg - kg~ (-1) AFB1 dose mix material, continuously fed contaminated feed 28 d can be successfully established by AFB1 AA broilers sub chronic liver injury model; curcumin can effectively Antagonizing the subchronic liver injury induced by AFB1 in AA broilers, it plays a protective role in liver. Especially, the protective effect of high dose curcumin is more durable, which can effectively alleviate the toxicity of AFB1. It is suggested that the dosage of curcumin in mixed feeding is 450 mg kg~ (-1).
