【摘要】：將18頭健康成年中國(guó)荷斯坦奶牛隨機(jī)分成高、中、低3組,每組每頭分別肌肉注射氨基丁三醇前列腺素F2α注射液10(50mg),5(25mg),2.5mL(12.5mg),采用酶聯(lián)免疫吸附法(ELISA)測(cè)定血藥濃度,用殘數(shù)法逐頭奶牛擬合藥動(dòng)學(xué)曲線方程并計(jì)算藥動(dòng)學(xué)參數(shù),研究氨基丁三醇前列腺素F2α在奶牛體內(nèi)的藥代動(dòng)力學(xué)。結(jié)果表明,藥時(shí)數(shù)據(jù)符合有吸收一室模型,高,中,低組的主要藥動(dòng)學(xué)參數(shù):吸收速率常數(shù)Ka分別為(18.22±2.17),(15.30±0.64),(20.09±5.03)h-1;吸收半衰期t1/2Ka分別為(0.04±0.004),(0.05±0.002),(0.04±0.01)h;分布和消除速率常數(shù)Ke分別為(1.32±0.14),(1.27±0.12),(1.40±0.16)h-1;消除半衰期t1/2Ke分別為(0.53±0.06),(0.55±0.05),(0.50±0.06)h;達(dá)峰時(shí)間tmax均為0.17h;達(dá)峰質(zhì)量濃度Cmax分別為(6.34±0.59),(5.94±0.27),(4.81±0.16)μg/L;藥時(shí)曲線下面積AUC0~t分別為(14.40±2.19),(10.14±1.07),(13.15±4.34)μg/(L·h)。奶牛單劑量肌肉注射氨基丁三醇前列腺素F2α注射液后,機(jī)體對(duì)藥物吸收迅速,消除快,濃度約10min達(dá)到峰值,100min后達(dá)到注射前的水平。根據(jù)藥時(shí)曲線峰濃度,AUC值及相關(guān)的臨床藥效學(xué)試驗(yàn)結(jié)果,建議臨床使用劑量為5mL(25mg)。
[Abstract]:Eighteen healthy adult Chinese Holstein cows were randomly divided into 3 groups: high, middle and low. Each group was given intramuscular injection of ABA prostaglandin F _ 2 偽 injection 10 ~ 50 mg / L ~ 2.5 mL / L ~ 12.5 mg 路g ~ (-1) 路L ~ (-1) 路L ~ (2.5) mg 路L ~ (-1) ~ (-1) 路L ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1)). The pharmacokinetics of aminobutanol prostaglandin F 2 偽 in dairy cows was studied by fitting the pharmacokinetic curve equation and calculating the pharmacokinetic parameters with the residue method. The results showed that the pharmacokinetics of ABA prostaglandin F 2 偽 was consistent with the absorption one-compartment model. The main pharmacokinetic parameters of the low group were as follows: the absorption rate constant Ka was 18.22 鹵2.17, the absorption half-life t1/2Ka was 0.04 鹵0.004, the absorption rate constant K _ e was 1.32 鹵0.14 and 1.27 鹵0.12, respectively; the half-life t1/2Ke was 0.53 鹵0.06, 0.55 鹵0.06, respectively; the peak time was 0.17h. The Cmax was 6.34 鹵0.59 渭 g / L and the area under the curve was 10.14 鹵1.07 鹵1.07 渭 g / L, respectively. After single dose injection of prostaglandin F _ 2 偽, aminobutanol prostaglandin F _ 2 偽 was injected intramuscularly in dairy cows. The concentration of 10min reached the level before injection. According to the peak concentration of AUC and the results of clinical pharmacodynamics test, it was suggested that the clinical dosage should be 5 mL / L ~ 25 mg / g ~ (-1) ~ (-1) 路min ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1) ~ (-1)).
