原花青素對(duì)玉米赤霉烯酮誘導(dǎo)小鼠睪丸TM4支持細(xì)胞凋亡的保護(hù)作用

【摘要】:為了研究原花青素對(duì)玉米赤霉烯酮誘導(dǎo)的小鼠睪丸支持細(xì)胞TM4凋亡的保護(hù)作用,試驗(yàn)采用體外培養(yǎng)小鼠睪丸支持細(xì)胞TM4,用MTT法分別測(cè)定原花青素(PC)、玉米赤霉烯酮(ZEA)對(duì)TM4細(xì)胞生長(zhǎng)的影響,流式細(xì)胞術(shù)檢測(cè)PC對(duì)ZEA誘導(dǎo)的TM4細(xì)胞凋亡的保護(hù)作用,并檢測(cè)PC、ZEA引起的總超氧化物歧化酶(T-SOD)的活性變化及丙二醛(MDA)的含量變化。結(jié)果表明:PC可顯著增加TM4細(xì)胞的存活率,抑制TM4細(xì)胞的凋亡,提高T-SOD活性,降低MDA含量;有效抑制ZEA誘導(dǎo)的TM4細(xì)胞凋亡,主要是通過(guò)抗氧化作用對(duì)經(jīng)ZEA染毒的TM4細(xì)胞起到保護(hù)作用。
[Abstract]:To study the protective effect of proanthocyanidins on TM4 apoptosis induced by zearalenone in mouse testicular Sertoli cells. Mouse testicular Sertoli cells (TM4) were cultured in vitro. The effects of proanthocyanidin and zeatin on the growth of TM4 cells were determined by MTT assay. The protective effects of PC on ZEA induced apoptosis of TM4 cells were detected by flow cytometry. The activity of total superoxide dismutase (T-SOD) and the content of malondialdehyde (MDA) were measured. The results showed that TM4 cells were significantly increased by TM4, apoptosis of TM4 cells was inhibited, the activity of T-SOD was increased, and the content of MDA was decreased. The effective inhibition of TM4 cell apoptosis induced by ZEA is mainly due to the protective effect of antioxidation on TM4 cells exposed to ZEA.
