【摘要】:膽汁酸(blie acid)是動(dòng)物體內(nèi)膽汁中的主要成分,直接參與體內(nèi)脂類(lèi)代謝過(guò)程。;蛆Z去氧膽酸(Taurochenodeoxycholic Acid,TCDCA)是膽汁酸中主要的生物活性物質(zhì),主要存在于雞膽汁中,具有抗氧化、解熱、抗炎、免疫、鎮(zhèn)痛等多種生物功能。為了深入了解TCDCA的生物活性,本課題針對(duì)TCDCA對(duì)脂肪代謝的影響進(jìn)行了試驗(yàn)研究。研究?jī)?nèi)容包括:經(jīng)口服給予不同劑量的TCDCA 21d后,采用全自動(dòng)生化分析儀檢測(cè)TCDCA對(duì)大鼠脂肪代謝的相關(guān)生理生化指標(biāo);分別采用TBA法、WSF-1法和分光光度法測(cè)定血清中丙二醛(MDA)含量、超氧化物歧化酶(SOD)和谷胱甘肽過(guò)氧化物酶(GSH-Px)活力;采用組織病理學(xué)方法檢測(cè)大鼠肝臟組織形態(tài)和細(xì)胞結(jié)構(gòu);采用免疫組化方法檢測(cè)法尼醇X受體(FXR)在大鼠肝臟的表達(dá)位置;分別采用熒光定量PCR和Western blot技術(shù)檢測(cè)大鼠肝臟中FXR基因、蛋白的表達(dá)量。研究結(jié)果表明:中劑量組的大鼠體重及增重均顯著低于陰性對(duì)照組(P<0.05);低、中、高三個(gè)劑量組肝體比極顯著大于陰性對(duì)照組(P<0.01);與陰性對(duì)照組相比,TCDCA低、中、高三個(gè)劑量組的TC、TG、HDL-C,LDL-C水平、腎體比、體脂肪率、料肉比以及各項(xiàng)血液生理生化指標(biāo)均無(wú)顯著性差異(P>0.05);中劑量TCDCA可顯著性降低大鼠血清中MDA含量,TCDCA處理組的大鼠血清中S0D和GSH-Px活力雖有升高,但差異不顯著(P>0.05);TCDCA給藥組的大鼠肝臟與其正常組織形態(tài)結(jié)構(gòu)相比均無(wú)顯著變化(P>0.05);免疫組化結(jié)果顯示,FXR在大鼠肝臟細(xì)胞核上均有表達(dá),但TCDCA給藥組的大鼠肝臟中FXR的表達(dá)量較多;TCDCA低劑量組的大鼠肝臟中FXR mRNA表達(dá)量極顯著高于陰性對(duì)照組(P<0.01),Western blot結(jié)果顯示TCDCA低劑量組FXR蛋白表達(dá)量顯著高于陰性對(duì)照組(P<0.05)。結(jié)論:中劑量TCDCA顯著降低大鼠體質(zhì)量,顯著抑制大鼠增重;FXR在大鼠肝臟中有較高效表達(dá),且TCDCA處理組的大鼠肝臟中FXR表達(dá)量明顯高于陰性對(duì)照組:低劑量TCDCA能夠顯著性升高FXR的蛋白和基因的表達(dá)量。
[Abstract]:Taurochenodeoxycholic acid (Taurochenodeoxycholic acid TCDCAA) is the main bioactive substance in bile acid, which mainly exists in chicken bile. In order to understand the biological activity of TCDCA, the effects of TCDCA on fat metabolism were studied. The content of malondialdehyde (MDA) in serum was determined by TBA method and spectrophotometry, respectively, using automatic biochemical analyzer to detect the physiological and biochemical indexes related to fat metabolism of rats by TCDCA, and to determine the content of malondialdehyde (MDA) in serum by TBA method and spectrophotometry, respectively. The activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) were detected by histopathological method, and the expression of FXR in rat liver was detected by immunohistochemical method. Fluorescence quantitative PCR and Western blot were used to detect the expression of FXR gene and protein in the liver of rats. The results showed that the weight and weight gain of middle dose group were significantly lower than those of negative control group (P < 0.05, P < 0.05, P < 0.05, P < 0.05). The liver body ratio of the three dose groups was significantly higher than that of the negative control group (P < 0.01), and the TCDCA was lower than that of the negative control group. There was no significant difference in feed / meat ratio and blood physiological and biochemical indexes (P > 0.05), and moderate dose TCDCA could significantly decrease the content of MDA in serum and increase the activity of S0D and GSH-Px in serum of rats treated with TCDCA-treated group. However, there was no significant difference between the two groups (P > 0.05) and the expression of FXR was found in the nuclei of the liver of the rats treated with TCDCA (P > 0.05), but there was no significant difference between the two groups (P > 0.05). However, the expression of FXR mRNA in the liver of the TCDCA group was significantly higher than that of the negative control group (P < 0.01). The results showed that the expression of FXR protein in the low-dose TCDCA group was significantly higher than that in the negative control group. Conclusion: middle dose TCDCA can significantly reduce the body mass of rats. The expression of FXR in the liver of rats treated with TCDCA was significantly higher than that in the control group. Low dose TCDCA could significantly increase the expression of FXR protein and gene.
