綿羊繁殖性狀遺傳機理的研究

【摘要】:解析綿羊繁殖性狀的遺傳機理是增加產(chǎn)羔數(shù)和提高養(yǎng)羊生產(chǎn)效率的重要基礎(chǔ)。自宋末蒙古羊南移以來,高繁的小尾寒羊和湖羊與低繁的蘇尼特羊和烏珠穆沁羊從原始群體演化至今已有上千年的歷史。在這一遺傳背景下,利用NGS技術(shù)篩選與綿羊繁殖力相關(guān)的遺傳變異成為可能。本研究通過比較四個蒙古羊后裔品種的基因組變異,并整合其它品種卵泡發(fā)育轉(zhuǎn)錄組和基因型數(shù)據(jù)初步解析了綿羊繁殖性狀的遺傳機理。主要研究結(jié)果如下:1、蒙古羊內(nèi)部分化研究通過滑動窗口計算ROH與Fst的方法,得到47個在高繁殖力群體與低繁殖力群體中發(fā)生明顯分化的區(qū)域,進一步功能分析找到6個參與到繁殖生物學過程的重要候選基因(如DACH2, AMH, ADAM21等)。2、表達相關(guān)性分析結(jié)合Miao等人發(fā)現(xiàn)的差異表達基因,通過表達相關(guān)性分析找到了4個與卵泡發(fā)育相關(guān)的關(guān)鍵基因(如ECH1, IGFIR等)及7個與卵泡發(fā)育相關(guān)的QTL。3、與繁殖性狀相關(guān)SNP標記篩選結(jié)合簡化基因組測序,利用關(guān)聯(lián)分析獲得了145個與綿羊繁殖性狀相關(guān)的多態(tài)位點。本研究首次通過多組學結(jié)合對綿羊繁殖性狀分子機理進行了探究,研究結(jié)果為后續(xù)功能基因的篩選及驗證提供了數(shù)據(jù)資源,并對定位綿羊繁殖性狀候選基因及育種工作奠定了基礎(chǔ)。
[Abstract]:The genetic mechanism of sheep reproductive traits is an important basis for increasing lambs and raising sheep productivity. It has been thousands of years since the high propagation of small tail Han sheep and Hu sheep and the low breeding Sunita sheep and Wuzhimuqin sheep evolved from the primitive population. It is possible to screen the genetic variation associated with sheep fecundity by using NGS technique. The genetic mechanism of sheep reproductive traits was preliminarily analyzed by integrating the data of follicular development transcriptome and genotype of other breeds. The main results were as follows: 1. The internal differentiation of Mongolian sheep was studied by sliding window method to calculate ROH and Fst. There were 47 areas with obvious differentiation between high fecundity population and low fecundity population. Further functional analysis found six important candidate genes involved in reproductive biology (such as DACH2, AMH, ADAM21, etc.). Expression correlation analysis combined with differentially expressed genes discovered by Miao et al. Four key genes related to follicular development (E. g. ECH1, IGFIR, etc.) and seven QTL.3related to follicular development were identified by expression correlation analysis. The results showed that QTL.3 was associated with the screening of SNP markers related to reproductive traits and simplified genomic sequencing. A total of 145 polymorphic loci related to sheep reproductive traits were obtained by association analysis. The results provided data resources for screening and verification of functional genes and laid a foundation for locating candidate genes for sheep reproductive traits and breeding.
