【摘要】：為了研究犬血型DEA1.1抗原生化特性,為犬輸血前血型檢測提供理論依據(jù),試驗采用凝聚胺介質(zhì)試劑調(diào)查356只不同地區(qū)及品種犬的DEA1.1血型頻率分布,用考馬斯亮藍(lán)法檢測DEA1.1紅細(xì)胞膜蛋白并通過免疫沉淀鑒定血型蛋白大小,觀察多種酶處理對DEA1.1抗原性的影響,通過ABO血型檢測試劑和ABO型微柱凝膠檢測卡檢測人類紅細(xì)胞ABO血型與犬DEA1.1血型有無交叉反應(yīng)。結(jié)果表明:不同品種犬的血型頻率差異較大,DEA1.1血型為50 ku和200 ku膜蛋白;DEA1.1抗原在無花果酶、木瓜酶、鏈霉蛋白酶處理后抗原性增強(qiáng),在菠蘿酶、糜蛋白酶和胰蛋白酶處理后抗原性未增強(qiáng);DEA1.1陰、陽性紅細(xì)胞與ABO血型試劑均未發(fā)生凝集。說明為降低輸血反應(yīng),最好采用DEA1.1血型頻率低的犬種作為供血犬,并建議輸血前采用木瓜酶介質(zhì)交叉配血試驗進(jìn)行血型檢測。
[Abstract]:In order to study the biochemical characteristics of canine blood group DEA1.1 antigen and provide theoretical basis for blood group detection before blood transfusion, the frequency distribution of DEA1.1 blood group in 356 dogs from different regions and breeds was investigated by polyacrylamide medium reagent. The erythrocyte membrane protein of DEA1.1 was detected by Coomassie brilliant blue method and the size of blood group protein was identified by immunoprecipitation. The effects of various enzyme treatments on the antigenicity of DEA1.1 were observed. ABO blood group detection reagent and ABO microcolumn gel test card were used to detect the cross reaction between human red blood cell ABO blood group and canine DEA1.1 blood group. The results showed that the frequency of DEA1.1 blood group was 50 ku and 200ku membrane. Protein DEA1.1 antigens in figs, The antigenicity of papain and strepsin was enhanced, but the antigenicity of Pineapple, chymotrypsin and trypsin was not enhanced after treatment with DEA1.1 negative, the agglutination of positive erythrocytes and ABO blood group reagent was not observed. The dogs with low DEA1.1 blood group frequency should be used as blood supplying dogs, and the papain enzyme medium cross matching test should be used to detect blood type before blood transfusion.
