豬ZP4基因的SNP鑒定、選擇壓及其與產(chǎn)仔數(shù)的關(guān)聯(lián)分析

【摘要】:本試驗(yàn)旨在研究豬ZP4基因的SNP在群體中的選擇壓及其與產(chǎn)仔數(shù)的關(guān)聯(lián)性。收集大白豬、長白豬、長大二元豬3個(gè)群體共計(jì)328頭母豬的產(chǎn)仔數(shù)記錄,利用直接測(cè)序法克隆ZP4基因中的SNP位點(diǎn),采用廣義顯性模型分析ZP4基因SNP與產(chǎn)仔數(shù)的關(guān)聯(lián)性。采用lositan軟件,Tajima′s D檢驗(yàn)和Fu and Li′s D檢驗(yàn)來檢測(cè)ZP4的選擇壓。結(jié)果發(fā)現(xiàn),ZP4中共檢測(cè)到27個(gè)SNPs,關(guān)聯(lián)分析表明A268T在3個(gè)豬群體中均與總產(chǎn)仔數(shù)顯著關(guān)聯(lián)(P0.05),A268T在長白豬和大白豬群體中與產(chǎn)活仔數(shù)顯著關(guān)聯(lián)(P0.05),群體遺傳學(xué)分析表明,T2950C位點(diǎn)受到了正選擇作用,雖然該位點(diǎn)突變與產(chǎn)仔數(shù)關(guān)聯(lián)不顯著,但是T2950C與A268T存在緊密連鎖。關(guān)聯(lián)分析和選擇壓力分析說明ZP4基因可以作為豬產(chǎn)仔數(shù)的候選標(biāo)記。
[Abstract]:The purpose of this study was to study the selective pressure of SNP of ZP4 gene in pig population and its correlation with litter size. The SNP site of ZP4 gene was cloned by direct sequencing. The relationship between ZP4 gene SNP and litter size was analyzed by generalized dominant model. The selection pressure of ZP4 was detected by lositan software Tajimahe D test and Fu and Li's D test. A total of 27 SNPs were detected in ZP4, and the correlation analysis showed that A268T was 3. There was a significant correlation between P0.05A268T and total litter size in the whole population. The population genetic analysis showed that the locus of T2950C was positively selected in Landrace pigs and large white pigs, and there was a significant correlation between P0.05T and live litter size. Although there was no significant correlation between the mutation and litter size, T2950C was closely linked to A268T. Association analysis and selection pressure analysis showed that ZP4 gene could be used as a candidate marker for piglet litter size.
