檸檬苦素對牛前體脂肪細(xì)胞分化過程中核纖層蛋白基因表達(dá)的影響

【摘要】:核纖層位于核膜內(nèi)側(cè),主要由核纖層蛋白構(gòu)成,對細(xì)胞核起重要的機(jī)械支持作用。編碼核纖層蛋白的基因突變能導(dǎo)致人類肌肉、脂肪等組織病變,統(tǒng)稱為核纖層病。為了研究檸檬苦素對牛前體脂肪細(xì)胞分化過程中核纖層蛋白基因表達(dá)的影響,試驗利用qRT-PCR技術(shù)分析了編碼核纖層蛋白的LMNA、LMNB1和LMNB2基因在牛前體脂肪細(xì)胞誘導(dǎo)分化過程中的表達(dá)變化,同時在細(xì)胞培養(yǎng)基中添加檸檬苦素,以分析檸檬苦素對細(xì)胞分化和核纖層蛋白編碼基因表達(dá)的影響。結(jié)果表明:在培養(yǎng)基中添加檸檬苦素能夠有效抑制牛前體脂肪細(xì)胞向成熟脂肪細(xì)胞的分化,試驗組細(xì)胞中脂質(zhì)沉積量極顯著低于對照組(P0.01)。LMNA基因在牛前體脂肪細(xì)胞中具有較高的表達(dá)水平,添加檸檬苦素的試驗組LMNA基因表達(dá)水平有降低的趨勢。試驗組LMNB1和LMNB2基因表達(dá)量在誘導(dǎo)分化的第0.5天極顯著低于對照組(P0.01),在誘導(dǎo)分化的10~12 d試驗組LMNB1基因表達(dá)量顯著高于對照組(P0.05)。LMNA、LMNB1和LMNB2基因在牛前體脂肪細(xì)胞誘導(dǎo)分化過程中具有相似的表達(dá)變化趨勢,均在誘導(dǎo)分化開始的0.5~1 d表達(dá)量升高。說明檸檬苦素在牛前體脂肪細(xì)胞早期分化時具有抑制核纖層蛋白基因表達(dá)的作用,而在分化后期則能夠促進(jìn)核纖層蛋白基因表達(dá)。
[Abstract]:The nuclear fiber layer is located on the inner side of the nuclear membrane and is mainly composed of nuclear laminin, which plays an important role in mechanically supporting the nucleus. The mutation of the gene encoding the nuclear laminin can lead to human muscle, fat and other tissue lesions. In order to study the effect of limonin on the expression of nuclear fiber laminin gene during the differentiation of bovine precursor adipocytes, QRT-PCR technique was used to analyze the expression changes of LMNAN LMNB1 and LMNB2 genes in bovine precursor adipocytes, and limonin was added to cell culture medium. In order to analyze the effect of limonin on cell differentiation and the expression of nuclear laminin coding gene, the results showed that the addition of limonin in culture medium could effectively inhibit the differentiation of bovine precursor adipocytes into mature adipocytes. The level of lipid deposition in the experimental group was significantly lower than that in the control group, and the expression level of LMNA gene in bovine precursor adipocytes was higher than that in the control group. The expression of LMNB1 and LMNB2 genes in the experimental group was significantly lower than that in the control group on the 0.5 day after differentiation, and the expression of LMNB1 gene in the experimental group was significantly lower than that in the control group on the 10th and 12th day after inducing differentiation. The expression of LMNB1 and LMNB2 genes in the preadipocytes was significantly higher than that in the control group. There was a similar trend in the expression of LMNB1 and LMNB2 in the differentiation of bovine preadipocytes. These results indicated that limonin could inhibit the expression of nuclear laminin gene in the early differentiation of bovine precursor adipocytes, while it could promote the expression of nuclear laminin gene at the late stage of differentiation.
