MUC4和MUC13基因在ETECF18抗性型和易感型梅山斷奶仔豬間的差異表達(dá)分析

【摘要】:MUC4和MUC13基因作為產(chǎn)腸毒素大腸桿菌(enterotoxigenic Escherichia coil,ETEC)F4的重要抗性候選基因,可能在斷奶仔豬抗ETEC F18菌株的感染過(guò)程中發(fā)揮重要作用,因此本試驗(yàn)運(yùn)用實(shí)時(shí)熒光定量PCR法分析了MUC4和MUC13基因在梅山豬ETEC F18抗性型和易感型個(gè)體11個(gè)組織(心臟、肝臟、脾臟、肺臟、腎臟、胃、肌肉、胸腺、淋巴結(jié)、十二指腸和空腸)中的表達(dá)水平及差異。結(jié)果顯示,MUC4和MUC13基因在檢測(cè)的11個(gè)組織中均有不同程度的表達(dá)。其中,在胸腺和淋巴組織中,MUC4基因在抗性型個(gè)體中的表達(dá)水平顯著高于易感型個(gè)體(P0.05);在肺臟組織中,MUC13基因在抗性型個(gè)體中的表達(dá)水平顯著高于易感型個(gè)體(P0.05),且MUC13基因在抗性群體腸道中的表達(dá)水平較高。由此推測(cè),MUC4基因在免疫組織、MUC13基因在腸道組織的高表達(dá)可在一定程度上提高了機(jī)體的免疫能力,保護(hù)和潤(rùn)滑腸道,從而抵抗ETEC F18菌株對(duì)機(jī)體的感染。
[Abstract]:MUC4 and MUC13 genes, which are important candidate genes for enterotoxigenic Escherichia Coilogenic Escherichia ETECF4, may play an important role in the process of weaning piglets resistant to ETEC F18. Therefore, real-time fluorescence quantitative PCR was used to analyze 11 tissues (heart, liver, spleen, lung, kidney, stomach, muscle, thymus, lymph node) of ETEC F18 resistant and susceptible individuals of Meishan pig. The expression levels and differences in duodenum and jejunum. The results showed that MUC4 and MUC13 genes were expressed to different degrees in 11 tissues. The expression level of MUC4 gene in thymocytes and lymphoid tissues was significantly higher than that in susceptible individuals (P 0.05), and in lung tissues was significantly higher than that in susceptible individuals (P 0.05), and the expression of MUC13 gene in susceptible individuals was significantly higher than that in susceptible individuals. The high expression of MUC4 gene in the immune tissue could improve the immune ability of the body to some extent, so the high expression of MUC13 gene in the intestinal tissue of the resistant population was higher than that of the control group, which suggested that the high expression of MUC4 gene in the immune tissue could improve the immune ability of the body. Protect and lubricate the intestines against ETEC F 18 infection.
