絨山羊胎兒成纖維細(xì)胞和成體干細(xì)胞體外生長特性的比較

【摘要】:本研究旨在探索阿爾巴斯白絨山羊成體干細(xì)胞和胎兒成纖維細(xì)胞體外生長、增殖能力和胚胎干細(xì)胞特性的差異,為成體干細(xì)胞的應(yīng)用提供新的理論基礎(chǔ)。研究以胎兒成纖維細(xì)胞(fetal fibroblast cells,FFCs)作為對照,檢測阿爾巴斯白絨山羊3種成體干細(xì)胞,脂肪間充質(zhì)干細(xì)胞(adipose-derived stem cells,ADSCs)、骨髓間充質(zhì)干細(xì)胞(bone marrow stromal cells,BMSCs)和肌肉衛(wèi)星細(xì)胞(muscle-derived satellite cells,MDSCs)中多能性(NANOG,OCT4,SOX2)、增殖(TERT,PCNA)、凋亡(P53,BAX)基因的轉(zhuǎn)錄和表達(dá)。表明g ADSCs、g BMSCs和g MDSCs低表達(dá)NANOG、OCT4和SOX2,與g FFCs沒有明顯差異;TERT在g BMSCs和g MDSCs中的表達(dá)量高于g FFCs中的表達(dá)量,而在g ADSCs中的表達(dá)量則低于g FFCs;PCNA和BAX在g FFCs和g MDSCs中高表達(dá),在g ADSCs和g BMSCs中低表達(dá);P53在g BMSCs中的相對表達(dá)量較高,在g FFCs、g ADSCs和g MDSCs中的相對表達(dá)量均較低。現(xiàn)象表明,在一定程度上成體干細(xì)胞的胚胎干細(xì)胞特性及增殖能力優(yōu)于胎兒成纖維細(xì)胞。
[Abstract]:The aim of this study was to explore the difference between adult stem cells and fetal fibroblasts in vitro growth, proliferation and characteristics of embryonic stem cells. To provide a new theoretical basis for the application of adult stem cells, fetal fibroblasts (FFCs) were used as control. Three kinds of adult stem cells and adipose-derived stem cells were detected. Bone marrow stromal cells. BMSCs) and muscle-derived satellite cells (MDSCs). The transcription and expression of SOX2, ADSCs, P53 and Bax) genes showed that g ADSCs. G BMSCs and g MDSCs were not significantly different from g FFCs in the expression of NANOGN OCT4 and SOX2. The expression of TERT in g BMSCs and g MDSCs was higher than that in g FFCs, but the expression in g ADSCs was lower than that in g FFCs. The expression of PCNA and BAX was high in g FFCs and g MDSCs, and low in g ADSCs and g BMSCs. The relative expression of p53 in g BMSCs was higher than that in g FFCsG ADSCs and g MDSCs. To a certain extent, the embryonic stem cells of adult stem cells are superior to fetal fibroblasts in the characteristics and proliferation of embryonic stem cells.
