深縣豬保種群遺傳多樣性簡(jiǎn)化監(jiān)測(cè)方法研究

【摘要】:為建立深縣豬種群多樣性監(jiān)測(cè)的簡(jiǎn)化指標(biāo)體系,試驗(yàn)采集河北省深縣豬4個(gè)世代179頭豬樣本,采用聯(lián)合國(guó)糧農(nóng)組織和國(guó)際動(dòng)物遺傳協(xié)會(huì)聯(lián)合推薦的27個(gè)微衛(wèi)星標(biāo)記進(jìn)行群體遺傳多樣性分析,對(duì)27個(gè)微衛(wèi)星標(biāo)記進(jìn)行兩階段、多梯度抽樣,并根據(jù)抽樣群體和總?cè)后w的多態(tài)信息含量(PIC)值的變化確定適宜抽樣微衛(wèi)星位點(diǎn)數(shù),再以4個(gè)世代樣本進(jìn)行驗(yàn)證。結(jié)果表明,選取8個(gè)微衛(wèi)星位點(diǎn)時(shí)與全部位點(diǎn)的比較相對(duì)偏差可控制在5%以?xún)?nèi),且以各分世代的驗(yàn)證比較相對(duì)偏差在10%左右,通過(guò)比較確定以S0155、S0228、SW632、S0090、CGA、S0101、S0227、SW122這8個(gè)微衛(wèi)星位點(diǎn)構(gòu)成的簡(jiǎn)化指標(biāo)體系可較好地反映深縣豬種群遺傳多樣性的變化。因此,深縣豬的保種群群體遺傳多樣性監(jiān)測(cè)可以簡(jiǎn)化為檢測(cè)這8個(gè)微衛(wèi)星位點(diǎn),同時(shí)本研究采用的方法也可為其他保種群的遺傳多樣性監(jiān)測(cè)方法的簡(jiǎn)化處理提供參考。
[Abstract]:In order to establish a simplified index system for monitoring the population diversity of pigs in Shenxian County, 179 pig samples from 4 generations of pigs in Shenxian County, Hebei Province were collected. The 27 microsatellite markers recommended jointly by FAO and the International Association of Animal Genetics were used for population genetic diversity analysis, and 27 microsatellite markers were sampled in two stages with multi-gradient sampling. According to the variation of the polymorphic information content (PICs) of the sampling population and the total population, the suitable number of microsatellite sites for sampling was determined, and the results were verified by four generations of samples. The relative deviation between 8 microsatellite loci and all the loci can be controlled within 5%, and the relative deviation of comparison between each generation is about 10%, and S0155 is determined by comparison. S0228,SW632,S0090,CGA,S0101,S0227. The simplified index system composed of 8 microsatellite loci of SW122 can well reflect the variation of genetic diversity of pig population in Shenxian county. The genetic diversity monitoring of preserved populations in Shenxian pig population can be simplified to detect the 8 microsatellite loci, and the method used in this study can also provide a reference for the simplification of genetic diversity monitoring methods of other protected populations.
