茶皂素對PRRSV感染Marc-145細胞受體表達及Caspase-9活化的影響

【摘要】:目的:以茶皂素(Tea saponin, TS)作為候選藥物,通過研究茶皂素對Marc-145細胞受體CD163和波形蛋白(Vimentin)基因合成和蛋白表達是否有影響,以及茶皂素是否能通過細胞凋亡內(nèi)源性通路影響PRRSV感染細胞,初步探究茶皂素的抗病毒作用機制。方法:通過qRT-PCR技術(shù)研究TS對細胞受體CD163和Vimentin基因合成的影響。同時運用Western blot技術(shù)檢測TS對細胞受體CD163和Vimentin蛋白表達量的影響。另外,通過Western blot技術(shù)研究TS對細胞內(nèi)源性凋亡通路啟動子Caspase-9活化的影響,初探TS的抗病毒機制。結(jié)果:1.qRT-PCR結(jié)果表明TS能顯著抑制感染PRRSV的Marc-145細胞受體CD163和Vimentin基因的合成。2.Western blot結(jié)果表明TS能顯著抑制細胞受體CD163和Vimentin的蛋白表達。3.Western blot結(jié)果表明TS能夠引起細胞內(nèi)源性凋亡通路啟動子Caspase-9的活化。結(jié)論:1.TS能抑制PRRSV在Marc-145細胞上的穿入過程,從而達到抗PRRSV的作用。2.TS可能通過激活細胞凋亡內(nèi)源性通路以早期促進細胞凋亡的方式,而產(chǎn)生抗PRRSV的作用。
[Abstract]:Objective: to use tea saponin (TSX) as a drug candidate. The effects of tea saponin on gene synthesis and protein expression of Marc-145 receptor CD163 and vimentin were studied. And whether tea saponin can affect PRRSV infected cells through the endogenous pathway of apoptosis. To explore the antiviral mechanism of tea saponin. Methods:. The effects of TS on the synthesis of CD163 and Vimentin genes were studied by qRT-PCR. Meanwhile, Western was used to study the effect of TS on the synthesis of CD163 and Vimentin genes. Blot technique was used to detect the effect of TS on the expression of CD163 and Vimentin protein. The effects of TS on the activation of Caspase-9 promoter of intracellular apoptotic pathway were studied by Western blot technique. Preliminary study on antiviral mechanism of TS. Results:. 1. The results of qRT-PCR showed that TS could significantly inhibit the synthesis of CD163 and Vimentin genes of Marc-145 cells infected with PRRSV. The results of blot showed that TS could significantly inhibit the protein expression of CD163 and Vimentin. 3. The results of blot showed that TS could induce the activation of Caspase-9 in the endogenous apoptotic pathway. 1. TS could inhibit the penetration of PRRSV in Marc-145 cells. TS may promote apoptosis by activating endogenous pathway of apoptosis, which may have the effect of anti-#en1#. 2.
