Forskolin引起大鼠脂肪細(xì)胞內(nèi)脂滴重塑(英文)

脂肪組織是人體的主要能量?jī)?chǔ)庫(kù),甘油三酯貯存在細(xì)胞內(nèi)脂滴(lipid droplets, LDs)中,越來(lái)越多的研究表明脂滴是細(xì)胞內(nèi)代謝活躍的細(xì)胞器。本研究旨在探討forskolin長(zhǎng)時(shí)間刺激脂肪分解過(guò)程中脂滴形態(tài)和脂滴表面perilipin家族蛋白的改變。以Sprague-Dawley (SD)大鼠附睪脂肪墊來(lái)源的分化脂肪細(xì)胞為研究對(duì)象,給予1μmol/L forskolin慢性刺激24 h,采用比色方法測(cè)定培養(yǎng)基中甘油的濃度;采用尼羅紅染色觀察細(xì)胞內(nèi)脂滴形態(tài)的變化;采用熒光定量PCR檢測(cè)perilipin家族蛋白mRNA水平的改變;采用免疫印跡和免疫熒光染色觀察蛋白水平以及蛋白的亞細(xì)胞定位。結(jié)果表明,1μmol/L forskolin孵育24 h可以持續(xù)刺激脂肪分解。伴隨著脂肪分解的進(jìn)行,細(xì)胞內(nèi)脂滴形態(tài)逐漸發(fā)生改變,細(xì)胞內(nèi)聚集存在的大脂滴逐漸減少,位于細(xì)胞周邊的小脂滴逐漸增加,最終細(xì)胞內(nèi)大脂滴全部消失,取而代之的是在細(xì)胞質(zhì)中彌散存在的微小脂滴。在脂肪分解過(guò)程中,perilipin家族蛋白水平也發(fā)生明顯變化。分化成熟的脂肪細(xì)胞幾乎沒(méi)有Plin2蛋白表達(dá),而forskolin慢性刺激可以...
