流式細(xì)胞術(shù)檢測(cè)NK細(xì)胞的細(xì)胞毒作用的兩種方法比較

目的:比較并優(yōu)化流式細(xì)胞術(shù)檢測(cè)人NK細(xì)胞體外細(xì)胞毒功能的方法。方法:采用Calcein-AM釋放法或CFSE/7-AAD法分別對(duì)靶細(xì)胞進(jìn)行標(biāo)記,按10∶1和20∶1效靶比與7例NK細(xì)胞在37℃5%CO2培養(yǎng)箱共培養(yǎng)4 h,流式細(xì)胞術(shù)測(cè)定兩種不同標(biāo)記方法的靶細(xì)胞死亡率。結(jié)果:各實(shí)驗(yàn)組中CFSE/7-AAD法檢出的靶細(xì)胞死亡率均高于Calcein-AM釋放法,在低效靶比、弱細(xì)胞毒作用條件下,差異具有統(tǒng)計(jì)學(xué)意義。細(xì)胞毒作用較弱時(shí),Calcein-AM釋放法平均熒強(qiáng)度變化不顯著、結(jié)果誤差較大;而CFSE/7-AAD法則能更加敏感地檢出靶細(xì)胞的死亡率。結(jié)論:CFSE/7-AAD法能夠特異、靈敏地檢出NK細(xì)胞的細(xì)胞毒活性,所獲得的結(jié)果更為可靠。
