用人骨髓間充質(zhì)干細胞誘導抗砷細胞

【摘要】:研究目的:用低劑量NaAsO_2長期誘導人骨髓間充質(zhì)干細胞(mesenchymal stem cells,MSCs),獲得抗砷細胞,探討機體抗砷機制。 研究方法:通過比較不同年齡人骨髓MSCs細胞倍增時間,細胞貼壁率等生物學性狀及細胞的砷耐受性,篩選抗砷種子細胞;選取胎兒骨髓MSCs,采用細胞抑制率在5%-10~時的NaAsO_2濃度(1μmol/L)為砷誘導濃度,同時設一組不加砷培養(yǎng)的胎兒骨髓MSCs作為同步對照,用噻唑蘭法(MTT)監(jiān)測細胞抗砷性是否產(chǎn)生;采用氫化物發(fā)生—原子熒光方法檢測細胞砷含量,二硫代二硝基苯甲酸直接顯色法,比較抗砷細胞與同步對照組胞內(nèi)GSH,GST等砷代謝指標的變化,驗證細胞是否獲得抗砷性,并初步探討了細胞產(chǎn)生抗砷能力的機制。 結(jié)果:不同年齡人骨髓MSCs在細胞形態(tài)、生長特性等方面是相似的,胎兒骨髓MSCs的擴增潛能明顯強于成人和老人骨髓MSCs。選擇胎兒骨髓MSCs作為抗砷種子細胞,經(jīng)過18周NaAsO_2誘導后,細胞對急性砷中毒的耐受性明顯提高,排砷能力增強,細胞內(nèi)GSH和GST活性增高。 結(jié)論:胎兒骨髓MSCs體外增殖分裂能力最強。人骨髓MSCs在低劑量NaAsO_2長期誘導下,能分化為具有抗砷能力的細胞。
