兼性胞內(nèi)寄生菌HLK1~T對人宿主細胞的感染和持續(xù)性寄生的研究

【摘要】:目前,病原菌仍是人類健康最大的威脅之一,比如急性呼吸道感染、腹瀉、肺結(jié)核等疾病都是由于病原菌的感染所致。而且,新的細菌感染性疾病的不斷出現(xiàn),以及大量致病菌的反復(fù)出現(xiàn)和耐藥性的增加,對公眾健康造成可怕的威脅。因此,對潛在病原菌的發(fā)現(xiàn)以及病原菌致病機理的研究一直是研究的熱點之一。 人紅白血病細胞系K562細胞被廣泛用于實驗室的研究。當(dāng)我們對K562細胞進行核型分析過程中,意外地發(fā)現(xiàn)K562細胞染色標(biāo)本中有桿狀細菌樣的結(jié)構(gòu)。為了排除該實驗中相關(guān)實驗試劑的環(huán)境細菌污染,我們嚴格控制了各種實驗試劑和實驗操作。然而,多次重復(fù)實驗仍得到以上結(jié)果。因此,我們推論該細胞中可能存在一種細胞內(nèi)生菌。將K562細胞、RPMI 1640完全培養(yǎng)基以及各種實驗用試劑分別涂至基礎(chǔ)血平板(5%綿羊血)上,置37℃孵箱中培養(yǎng)48~72小時,結(jié)果發(fā)現(xiàn)涂有K562細胞的平板上有菌落生成,而其它平板均無任何菌落形成。經(jīng)多次實驗證明,平板上形成的細菌集落確為K562細胞來源。只得注意的是,K562細胞能保持良好的生長狀態(tài)和增殖能力,而且細胞培養(yǎng)基中無明顯的細菌污染跡象。以上初步的實驗結(jié)果提示,寄生在K562細胞內(nèi)的細菌可能是一種新的細胞內(nèi)生菌,因為目前已明確的能寄生在人類細胞內(nèi)的胞內(nèi)寄生菌在細胞內(nèi)會大量繁殖,因此感染的細胞不能健康生長。由于該菌可能是一種新的胞內(nèi)寄生菌,就有必要明確該菌在細菌分類中的地位,以及該菌的基本的生物學(xué)特性。 第一部分:HLK1~T的分離和鑒定 目的:對細菌的分類鑒定是了解其生物學(xué)功能和特性的基礎(chǔ)。為明確HLK1~T的分類地位及其基本生物學(xué)特性,該研究從人紅白血病細胞系562細胞中分離獲得
