兼性胞內寄生菌HLK1~T對人宿主細胞的感染和持續(xù)性寄生的研究

【摘要】:目前,病原菌仍是人類健康最大的威脅之一,比如急性呼吸道感染、腹瀉、肺結核等疾病都是由于病原菌的感染所致。而且,新的細菌感染性疾病的不斷出現(xiàn),以及大量致病菌的反復出現(xiàn)和耐藥性的增加,對公眾健康造成可怕的威脅。因此,對潛在病原菌的發(fā)現(xiàn)以及病原菌致病機理的研究一直是研究的熱點之一。 人紅白血病細胞系K562細胞被廣泛用于實驗室的研究。當我們對K562細胞進行核型分析過程中,意外地發(fā)現(xiàn)K562細胞染色標本中有桿狀細菌樣的結構。為了排除該實驗中相關實驗試劑的環(huán)境細菌污染,我們嚴格控制了各種實驗試劑和實驗操作。然而,多次重復實驗仍得到以上結果。因此,我們推論該細胞中可能存在一種細胞內生菌。將K562細胞、RPMI 1640完全培養(yǎng)基以及各種實驗用試劑分別涂至基礎血平板(5%綿羊血)上,置37℃孵箱中培養(yǎng)48~72小時,結果發(fā)現(xiàn)涂有K562細胞的平板上有菌落生成,而其它平板均無任何菌落形成。經多次實驗證明,平板上形成的細菌集落確為K562細胞來源。只得注意的是,K562細胞能保持良好的生長狀態(tài)和增殖能力,而且細胞培養(yǎng)基中無明顯的細菌污染跡象。以上初步的實驗結果提示,寄生在K562細胞內的細菌可能是一種新的細胞內生菌,因為目前已明確的能寄生在人類細胞內的胞內寄生菌在細胞內會大量繁殖,因此感染的細胞不能健康生長。由于該菌可能是一種新的胞內寄生菌,就有必要明確該菌在細菌分類中的地位,以及該菌的基本的生物學特性。 第一部分:HLK1~T的分離和鑒定 目的:對細菌的分類鑒定是了解其生物學功能和特性的基礎。為明確HLK1~T的分類地位及其基本生物學特性,該研究從人紅白血病細胞系562細胞中分離獲得
