我國伯氏疏螺旋體分離株基因型及致病性研究

【摘要】:為了解我國伯氏疏螺旋體基因型分布,擴大我國伯氏疏螺旋體菌種庫,本研究選擇內(nèi)蒙古自治區(qū)、黑龍江省、吉林省、山東省、浙江省和福建省的林區(qū)為調(diào)查點,采集全溝硬蜱、嗜群血蜱和森林革蜱等6種蜱類,進行伯氏疏螺旋體分離和鑒定。結(jié)果僅從全溝硬蜱中分離到26株分離株。 采用RFLP、SSCP及測序方法對26株分離株進行基因分型,結(jié)果顯示5S-23S rRNA基因間隔區(qū)屬高變區(qū)中在Mse Ⅰ的識別序列中發(fā)生了堿基置換,導致RFLP方法不能對部分分離株進行明確分型;SSCP缺乏類似于GenBank的數(shù)據(jù)庫,不能對未知菌株進行準確分型;序列測定方法最為適用于分離株的基因型鑒定。采用該方法對本室現(xiàn)有我國36株分離株進行基因型分析,結(jié)果顯示這些分離株分別屬于3種基因型:B.garinii、B.afzelii及B.valaslana,其中B.garinii(29/36)為優(yōu)勢基因型,主要分布于內(nèi)蒙古、黑龍江及吉林;B.afzelii基因型均來自于黑龍江;B.valaslana基因型均來自于云南。結(jié)果揭示我國伯氏疏螺旋體分離株的基因型分布具有明顯的地域差異。 為研究我國優(yōu)勢基因型B.garinii型分離株的致病性,,本研究選用3株具有遺傳差異的B.garinii型分離株經(jīng)皮下重復接種BALB/c小鼠3次,于40天及87天后分批處死后采集臟器,采用HE染色及免疫組化方法研究其急、慢性病理改變及其抗原分布。結(jié)果顯示3株分離株均致使實驗小鼠的組織臟器發(fā)生不同程度的病理變化,并由急性病變向慢性病變轉(zhuǎn)化,這可能是由于病原體長期感染所引起的。本研究中觀察到3株不同分離株導致的同期病理改變不盡相同,與以往報道也不相一致,可能是由我國分離株的遺傳差異所導致的,其機理有待進一步研究。
[Abstract]:In order to understand the genotypic distribution of Borrelia burgdorfera in China and expand the species bank of Borrelia burgdorfera in China, the forest areas of Inner Mongolia Autonomous region, Heilongjiang Province, Jilin Province, Shandong Province, Zhejiang Province and Fujian Province were selected as investigation points. Six kinds of ticks, such as hard ticks, Haemaphysalis viridis and Euphorbia pulcherrima, were collected and identified. Results only 26 strains were isolated from the whole gully ticks.
