rhGH對(duì)大鼠腸缺血再灌注腸壁免疫細(xì)胞中P53基因表達(dá)的實(shí)驗(yàn)研究

【摘要】: 目的:通過研究腸缺血再灌注損傷后應(yīng)用rhGH對(duì)腸道免疫屏障中T淋巴細(xì)胞亞群和漿細(xì)胞中P53基因表達(dá)的影響,探討在腸缺血再灌注損傷后應(yīng)用rhGH對(duì)腸黏膜免疫屏障的作用機(jī)制。 方法:取Wistar大鼠60只(購(gòu)自山東大學(xué)醫(yī)學(xué)院動(dòng)物實(shí)驗(yàn)中心)雌雄各半,隨機(jī)分為缺血再灌注對(duì)照組和缺血再灌注實(shí)驗(yàn)組各30只,再按實(shí)驗(yàn)天數(shù)分為2天、4天、6天組各10只。實(shí)驗(yàn)觀察期間,,Wistar大鼠在溫度相對(duì)恒定(25℃±1.5℃),通風(fēng)的條件下觀察,并給與實(shí)驗(yàn)用動(dòng)物飼料喂養(yǎng),然后通過夾閉腸系膜前動(dòng)脈45分鐘,制造腸缺血再灌注模型,而引起腸道屏障功能損害的病理現(xiàn)象。實(shí)驗(yàn)組每天給與rhGH(1.33u/Kg/d),于造模后2、4、6天處死相應(yīng)的大鼠,取回腸末端腸壁制作石蠟標(biāo)本,采用免疫組化雙染色法觀察其黏膜固有層內(nèi)CD_8~+,CD_4~+,CD_3~+T淋巴細(xì)胞數(shù)及Ig A漿細(xì)胞中P53基因表達(dá)的情況,通過方差分析的統(tǒng)計(jì)方法,比較實(shí)驗(yàn)組與對(duì)照組P53基因表達(dá)的差異。 結(jié)果:(1)第4、6天CD4~+T淋巴細(xì)胞、CD3~+T淋巴細(xì)胞中P53基因表達(dá)對(duì)照組高于實(shí)驗(yàn)組(P<0.05、P<0.01);第2、6天IgA漿細(xì)胞中P53基因表達(dá)對(duì)照組高于實(shí)驗(yàn)組(P<0.05、P<0.01)。(2)實(shí)驗(yàn)組與對(duì)照組相比較,CD8~+T淋巴細(xì)胞中P53基因的表達(dá)第2、4、6天均無(wú)差異性(P>0.05)。(3)對(duì)照組CD_4~+T淋巴細(xì)胞中P53基因表達(dá)第6天比第2、4天高(P<0.05);CD_3~+T淋巴細(xì)胞中P53基因表達(dá)第6天比第2、4天高(P<0.01、P<0.05),第4天比第2天高(P<0.01);IgA漿細(xì)胞中P53基因表達(dá)第6天比第2、4天高(P<0.01、P<0.05);(4)實(shí)驗(yàn)組CD_8~+T淋巴細(xì)胞中P53基因的表達(dá)第4、6天高于第2天(P<0.05);CD_4~+T淋巴細(xì)胞中P53基因表達(dá)第4、6天低于第2天(P<0.05)。 結(jié)論:在腸黏膜屏障損傷時(shí),對(duì)照組腸壁免疫細(xì)胞中P53基因的表達(dá)隨著再灌注損傷的進(jìn)行而逐漸增加;應(yīng)用rhGH能夠明顯抑制腸壁免疫細(xì)胞凋亡基因P53的表達(dá),rhGH對(duì)腸道免疫屏障的調(diào)節(jié)作用可能是通過對(duì)P53基因的調(diào)控而實(shí)現(xiàn)的,應(yīng)用時(shí)機(jī)在損傷后第4天更合適。
