細(xì)胞外基質(zhì)糖基化對(duì)表皮干細(xì)胞生物學(xué)特性的影響

【摘要】: [目的]糖尿病是日常生活常見(jiàn)疾病之一,糖尿病下肢潰瘍是其并發(fā)癥之一,且經(jīng)久難愈,因此糖尿病難愈創(chuàng)面成為人們研究的熱點(diǎn)。表皮干細(xì)胞作為皮膚發(fā)生修復(fù)重建的關(guān)鍵源泉,糖尿病創(chuàng)面難愈是否與此有關(guān)呢?本實(shí)驗(yàn)通過(guò)探討細(xì)胞基質(zhì)糖基化對(duì)表皮干細(xì)胞黏附、增殖、凋亡等生物學(xué)行為的影響,進(jìn)一步尋找糖尿病創(chuàng)面難愈機(jī)制。 [方法]利用Ⅳ膠原的快速黏附法分離培養(yǎng)表皮干細(xì)胞。然后利用免疫細(xì)胞化學(xué)和免疫熒光化學(xué)分別對(duì)角蛋白19和β1整合素進(jìn)行鑒別。制作并鑒定糖基化Ⅳ膠原,將制備的表皮細(xì)胞懸液分別接種在包被有正常Ⅳ膠原和糖基化Ⅳ膠原(膠原糖基化7天,14天,21天)的6孔板上。15分鐘后,每個(gè)組取3個(gè)孔,洗去未黏附細(xì)胞,分別計(jì)算其黏附率并進(jìn)行比較;取第2代表皮干細(xì)胞,以5×10~3接種于預(yù)先包被有正常Ⅳ膠原和糖基化Ⅳ膠原的96孔板內(nèi),分別取培養(yǎng)3小時(shí)、2天、4天、6天、8天、10天、12天、14天細(xì)胞每天取5孔用MTT比色法測(cè)OD值,取其平均值并繪制生長(zhǎng)曲線圖;取第2代成對(duì)數(shù)生長(zhǎng)期的糖基化組和正常膠原組細(xì)胞,按200/孔接種6孔板,2周之后測(cè)定克隆生成率;表皮干細(xì)胞按1×10~6/孔接種于包被有正常膠原和糖基化膠原6孔板上,培養(yǎng)6天和10天后流式細(xì)胞儀檢測(cè)Annexin V Fitc比較兩者細(xì)胞凋亡情況。 [結(jié)果]培養(yǎng)出來(lái)的表皮干細(xì)胞通過(guò)鑒定角蛋白19和β1整合素均有陽(yáng)性表達(dá);Ⅳ膠原糖基化7天,糖基化Ⅳ膠原的熒光值為1.864±0.126,而此時(shí)正常未進(jìn)行糖基化的Ⅳ膠原熒光值為0.699±0.0831,糖基化14天,糖基化Ⅳ膠原的熒光值為3.051±0.112,而正常Ⅳ膠原熒光值為0.765±0.0927;糖基化21天,糖基化Ⅳ膠原的熒光值為4.416±0.1378,而正常Ⅳ膠原熒光值為0.8254±0.058;膠原在高糖下孵育7天時(shí),糖基化組與正常膠原組黏附率沒(méi)差異,P值>0.05;膠原在高糖下孵育14天時(shí)糖基化組比正常組黏附率低,P值<0.05,21天時(shí)糖基化組比正常組黏附率顯著降低,P值<0.01;可見(jiàn)隨著糖基化程度的加深,對(duì)表皮干細(xì)胞的黏附率逐漸降低;通過(guò)MTT繪制表皮干細(xì)胞在正常Ⅳ膠原和糖基化Ⅳ膠原上的生長(zhǎng)曲線,可以看出在4天前細(xì)胞增殖緩慢,兩者間沒(méi)有差異,6天后開(kāi)始出現(xiàn)差異,正常膠原組細(xì)胞增長(zhǎng)速度要快于糖基化組;表皮干細(xì)胞在正常Ⅳ膠原和糖基化Ⅳ膠原上的克隆生成率比較:糖基化組為(38.5±1.9)%,正常膠原組為(32.7±1.5)%,糖基化組克隆生成率低于正常組膠原組。P<0.05;表皮干細(xì)胞在正常Ⅳ膠原和糖基化Ⅳ膠原上的凋亡比較:細(xì)胞孵育6天時(shí),正常膠原組Annexin V陽(yáng)性細(xì)胞數(shù)為(10.7±0.9438)%,而糖基化膠原組為(16.22±0.7912)%;細(xì)胞孵育第10天時(shí)正常膠原組Annexin V陽(yáng)性細(xì)胞數(shù)為(4.66±0.7368)%,而糖基化膠原組為(8.266±0.8177)%。糖基化膠原組較正常膠原組Annexin V陽(yáng)性細(xì)胞數(shù)明顯增多。P<0.05 [結(jié)論]基質(zhì)糖基化抑制了表皮干細(xì)胞的黏附和增殖,促進(jìn)了表皮干細(xì)胞的凋亡,可能是糖尿病創(chuàng)面難愈機(jī)制之一。
