鈣激活鉀通道及ATP敏感性鉀通道在缺氧性肺血管收縮中的作用

【摘要】: 用半定量RT-PCR及Western blot方法觀察缺氧對(duì)培養(yǎng)的大鼠肺動(dòng)脈平滑肌細(xì)胞(pulmonary artery smooth muscle cells, PASMCs)上高電導(dǎo)鈣激活鉀通道(BKCa )及ATP敏感性鉀通道Kir6.1和SUR2B基因表達(dá)及蛋白水平的影響。結(jié)果:(1)培養(yǎng)的PASMCs在基因水平和蛋白水平均存在BKCa及Kir6.1和SUR2B的表達(dá)。(2)急性缺氧6h在基因水平和蛋白水平均能上調(diào)BKCa及Kir6.1和SUR2B的表達(dá)。慢性缺氧48h時(shí)BKCa的基因水平和蛋白水平均上調(diào),慢性缺氧72h時(shí)Kir6.1的基因水平和蛋白水平均上調(diào)。(3)慢性缺氧48h復(fù)氧12h再缺氧6h及慢性缺氧72h復(fù)氧12h再缺氧6h,BKCa及Kir6.1和SUR2B在基因水平和蛋白水平與正常組相比,均無(wú)顯著性差異。結(jié)論:(1)BKCa及Kir6.1和SUR2B的基因水平和蛋白水平的表達(dá)在不同的缺氧時(shí)間上調(diào),其可能在缺氧性肺血管收縮(hypoxic pulmonary vasoconstriction, HPV)中起到了負(fù)性調(diào)節(jié)作用,從而降低HPV。(2)慢性缺氧后再急性缺氧,BKCa及Kir6.1和SUR2B在基因水平和蛋白水平與正常組相比,均無(wú)顯著性差異,這可能是機(jī)體的習(xí)服機(jī)制之一。
[Abstract]:Semi-quantitative RT-PCR and Western blot methods were used to observe the effect of hypoxia on (pulmonary artery smooth muscle cells, of cultured rat pulmonary artery smooth muscle cells The effects of high conductance calcium activated potassium channel (BKCa) and ATP sensitive potassium channel Kir6.1 and SUR2B gene expression and protein level on PASMCs). Results: (1) the expression of BKCa, Kir6.1 and SUR2B were found in cultured PASMCs at both gene and protein levels. (2) the expression of BKCa, Kir6.1 and SUR2B were up-regulated at gene and protein levels for 6 h after acute hypoxia. The gene and protein levels of BKCa were up-regulated at 48 h of chronic hypoxia and up-regulated at 72 h of chronic hypoxia. (3) the gene and protein levels of Kir6.1 were up-regulated at 48 h of chronic hypoxia and 12 h of reoxygenation for 12 h and 12 h of hypoxia for 12 h and 72 h of hypoxia for 12 h and 6 h, respectively. There was no significant difference in gene level and protein level of BKCa, Kir6.1 and SUR2B between normal group and control group. Conclusion: (1) the gene and protein levels of BKCa, Kir6.1 and SUR2B are up-regulated at different hypoxia time, which may play a negative role in hypoxic pulmonary vasoconstriction (hypoxic pulmonary vasoconstriction, HPV). There was no significant difference in gene and protein levels of BKCa, Kir6.1 and SUR2B between the two groups, which may be one of the mechanisms of acclimatization of HPV. (2).
