【摘要】:研究背景: 肺動(dòng)脈高壓是肺循環(huán)障礙和肺功能異常性疾病的常見表現(xiàn),是肺心病發(fā)病機(jī)制的中心環(huán)節(jié)。急性和慢性低氧均可導(dǎo)致缺氧性肺動(dòng)脈高壓,其中肺血管收縮反應(yīng)增強(qiáng)和肺血管結(jié)構(gòu)改建是其主要血管變化特征。肺動(dòng)脈平滑肌細(xì)胞(PASMC)的肥大增生是肺血管結(jié)構(gòu)改建的主要表現(xiàn)之一,缺氧性PASMC增生是肺動(dòng)脈高壓形成的重要原因之一。 降鈣素基因相關(guān)肽(Calcitonin gene-related peptide,CGRP)具有強(qiáng)大的舒血管作用,有研究報(bào)道缺氧性肺動(dòng)脈高壓大鼠的血漿CGRP水平明顯下降,,且肺動(dòng)脈壓力與血漿CGRP濃度呈顯著負(fù)相關(guān),而給大鼠注射CGRP,則可減輕或逆轉(zhuǎn)缺氧性肺動(dòng)脈高壓。 Rho GTP酶是真核細(xì)胞內(nèi)重要的信號(hào)傳遞分子,在真核細(xì)胞信號(hào)傳遞過程中發(fā)揮著分子開關(guān)作用。Rho激酶是RhoA的下游效應(yīng)蛋白,分子量約160kD,是一個(gè)Ser/Thr蛋白激酶。研究發(fā)現(xiàn)Rho激酶在凝血酶、血管緊張素Ⅱ、五羥色胺和Urotensin—Ⅱ等誘導(dǎo)的血管平滑肌細(xì)胞增生中
[Abstract]:Background: pulmonary hypertension is a common manifestation of pulmonary circulation disorder and abnormal pulmonary function, and is the central link of the pathogenesis of cor pulmonale. Both acute and chronic hypoxia can lead to hypoxic pulmonary hypertension, in which the enhancement of pulmonary vasoconstriction and the structural remodeling of pulmonary vessels are the main characteristics of pulmonary hypertension. Hypertrophic proliferation of pulmonary artery smooth muscle cells (PASMC) is one of the main manifestations of pulmonary vascular structural remodeling. Hypoxic PASMC proliferation is one of the important causes of pulmonary hypertension. Calcitonin gene-related peptide (Calcitonin gene-related peptide,CGRP) has a strong vasodilatory effect. It has been reported that the plasma CGRP level in hypoxic pulmonary hypertension rats is significantly decreased, and the pulmonary artery pressure is negatively correlated with the plasma CGRP concentration. Injection of CGRP, into rats could relieve or reverse hypoxic pulmonary hypertension. Rho GTP is an important signaling molecule in eukaryotic cells and plays a molecular switching role in eukaryotic signal transduction. Rho kinase is a downstream effector of RhoA with molecular weight of about 160kD.It is a Ser/Thr protein kinase.
