兩株甲真菌rDNAITS序列分析及系統(tǒng)形態(tài)學研究

【摘要】:甲真菌病是由真菌引起的一種慢性、頑固、防礙美容的常見傳染性疾病。本文對從甲病患者病甲中分離得到的菌株,進行個體發(fā)育學研究,彌補了當前甲真菌研究中對其分類地位確定的不完善之處,并探討了某些分子生物學技術在甲真菌分類中的應用價值。 1.通過體外模型實驗確定了菌株Fh_4和菌株Dh_3是引起甲部感染的病原真菌。 2.通過不同生長階段菌體的DNA提取結果,絲狀真菌DNA的提取應選在其對數(shù)生長期為好。 根據(jù)系統(tǒng)分類研究中的18S rDNA序列和rDNAITS序列(ITS1、5.8S rDNA、ITS2)與相關屬、種的序列構建的系統(tǒng)發(fā)育樹、序列相似性研究,菌株Fh_4的分類地位應是半知菌亞門(Fungi Imperfecfi)—球殼孢目(Sphaeropsidales)—球殼孢科(Sphaeropsidaceae)—莖點霉屬(Phoma)。菌株Dh_3的分類地位應是半知菌亞門(Fungi Imperfecti)—叢梗孢目(Moniliales)—叢梗孢科(Monilialiaceae)—枝頂孢屬(Acremonium)。 3.通過對菌株Fh_4和Dh_3進行了較為系統(tǒng)的形態(tài)學研究可知:菌株Fh_4的適宜的生長條件是28℃,pH 6.5-7.0;菌落從白色變?yōu)榧t色再變?yōu)榘迭S色;菌絲無色,具隔膜,分枝狀生長;能產生關節(jié)菌絲;分生孢子單胞紅色,橢圓形;分生孢子器球形,黑褐色,頂端有孔口,孢子成熟后從孔口逸出;沒有發(fā)現(xiàn)有性階段。菌株Dh_3適宜的生長條件是:28℃,pH 6.0-7.0;在察氏培養(yǎng)基上菌落淡紅色;營養(yǎng)菌絲匍匐生長,分枝、光滑、具隔膜,寬1.5μm,分生孢子梗散生,瓶梗直接從菌絲上長出,直立錐形,多單生,產孢瓶體細長28~38μm,圓柱狀,分生孢子單個循序產生,形成鏈狀,孢子橢圓形,表面光滑,4.4~5.2×1.4~2.0μm,未見厚垣孢子;未發(fā)現(xiàn)Dh_3產生有性孢子器。 4.通過以上研究及相關資料和檢索表可知:菌株Dh_3應是枝頂霉屬中的Acremonium egyptiacum菌株Fh_4屬于莖點霉屬,由于莖點霉屬菌種以前都是根據(jù)真菌感染的植物而命名,使得屬中的同種異名現(xiàn)象很嚴重,而且到現(xiàn)在未見有關莖點霉屬的真菌能引起甲真菌病的報道,建議給菌株Fh_4新的名稱。
[Abstract]:Onychomycosis is a chronic, persistent, beauty-impaired, common infectious disease caused by fungi. In this paper, the individual development of strains isolated from patients with nail disease was studied, which made up for the imperfections in the classification status of onychomycetes in the current research. The application value of some molecular biological techniques in the classification of onychomycetes was also discussed. 1. Strain Fh_4 and strain Dh_3 were identified as pathogenic fungi causing infection of part A by model experiment in vitro. 2. The extraction of DNA from filamentous fungi by DNA extraction at different growth stages should be selected in the logarithmic growth period of filamentous fungi. Based on the phylogenetic tree and sequence similarity of 18s rDNA sequence and rDNAITS sequence (ITS1,5.8S rDNA,ITS2) and related genera and species in phylogenetic classification, The taxonomic status of strain Fh_4 should be (Fungi Imperfecfi)-(Sphaeropsidales)-(Sphaeropsidaceae)-(Phoma). The taxonomic status of strain Dh_3 should be (Fungi Imperfecti)-(Moniliales)-(Monilialiaceae)-(Acremonium). 3. The morphology of strain Fh_4 and Dh_3 was studied systematically.
