層粘連蛋白對人絨毛膜促性腺激素刺激下卵巢顆粒細胞孕酮分泌的影響

【摘要】: 目的:研究層粘連蛋白(LN)對人絨毛膜促性腺激素(hCG)刺激下大鼠排卵前顆粒細胞孕酮(P)分泌的影響及可能的作用機制。 方法:分離未成熟大鼠排卵前顆粒細胞,分3組進行培養(yǎng):LN鋪板組、未鋪板組及LN鋪板同時加入抗整合素α_6單抗(GoH3)組。觀察比較細胞貼壁的情況。血清培養(yǎng)48h后加入hCG,無血清條件下再培養(yǎng)48h后收集培養(yǎng)液作孕酮測定及LH/hCG受體分析。 結(jié)果: LN鋪板組顆粒細胞6小時即可貼壁及展開,而未鋪板組16h開始貼壁及展開;無論hCG加入與否LN均可顯著刺激顆粒細胞孕酮的產(chǎn)生, 5ug/mL的GoH3能明顯抑制LN的此刺激作用。試驗還觀察到LN使顆粒細胞LH/hCG受體親和力升高,受體數(shù)目上調(diào),增強hCG刺激下顆粒細胞的孕酮分泌。 結(jié)論: LN通過與細胞表面粘附分子整合素α_6β_1(integrinα_6β_1)相互作用,可能為卵巢內(nèi)的一種局部調(diào)節(jié)因素,干擾LH/hCG受體功能并增強顆粒細胞孕酮的產(chǎn)生,在圍排卵期顆粒細胞黃素化過程中發(fā)揮重要效應(yīng)。
[Abstract]:Aim: to study the effect of laminin (LN) on progesterone (P) secretion in rat granulosa cells stimulated by human chorionic gonadotropin (hCG) and its possible mechanism. Methods: pre-ovulatory granulosa cells were isolated from immature rats and cultured in three groups: LN group, LN group and anti-integrin 偽 _ 6 monoclonal antibody (GoH3) group. Observe and compare the cell adhesion. After 48 hours of serum culture, hCG, was added to the serum-free medium for 48 h and then collected for progesterone determination and LH/hCG receptor analysis. Results: the granulosa cells in the LN laminate group could adhere and expand in 6 hours, while those in the non-laminate group began to adhere and expand at 16 h. Whether hCG was added or not, LN could significantly stimulate progesterone production in granulosa cells, and GoH3 of 5ug/mL could significantly inhibit the stimulation of LN. It was also observed that LN increased the affinity of LH/hCG receptors and increased the number of receptors in granulosa cells, and enhanced the progesterone secretion of granulosa cells stimulated by hCG. Conclusion: through the interaction of LN with integrin 偽 _ 6 尾 _ 1 (integrin 偽 _ 6 尾 _ 1), it may be a local regulatory factor in ovary, interfere with the function of LH/hCG receptor and enhance the production of progesterone in granulosa cells. It plays an important role in the process of luteinization of granulosa cells during the peri-ovulatory period.
