多巴胺受體D5B轉(zhuǎn)基因大鼠的建立及行為學(xué)研究

【摘要】:我們克隆得到了人類多巴胺受體D5基因的一個(gè)新亞型D5B。D5B基因的序列與D5有98%同源性,與D5的兩個(gè)假基因有94%同源性。D5B基因與假基因不同,含有一個(gè)完整的開放閱讀框,能夠編碼比D5受體多一個(gè)氨基酸的蛋白。藥理學(xué)研究初步表明D5B受體與D5受體的藥理學(xué)特性不同。為了研究D5B受體的功能,我們建立了由CamKⅡ啟動(dòng)子驅(qū)動(dòng)的前腦過表達(dá)D5B的轉(zhuǎn)基因大鼠。RT-PCR和Western blot分析表明D5B基因在轉(zhuǎn)基因大鼠腦部特異性表達(dá)。隨后運(yùn)用行為學(xué)方法對(duì)D5B轉(zhuǎn)基因大鼠進(jìn)行了焦慮、抑郁、學(xué)習(xí)和記憶方面的表型研究,結(jié)果表明D5B基因在許多大腦功能如情緒、學(xué)習(xí)和記憶方面起到復(fù)雜的調(diào)控作用。 1.D5B基因克隆的確證 在之前的研究中,我們是用Human Genomic DNA做模板,PCR得到的D5B基因。為了驗(yàn)證克隆的可靠性,我們分別以SH-Sy5Y和HEK293兩種細(xì)胞提取的基因組DNA作模板進(jìn)行PCR反應(yīng),之后將PCR產(chǎn)物克隆到T載體并測序。由測序結(jié)果分析,我們從42個(gè)測序克隆中得到12個(gè)含有D5B基因的克隆。 2.建立轉(zhuǎn)基因大鼠 大鼠作為一種模式動(dòng)物已經(jīng)被廣泛的應(yīng)用于藥理學(xué)研究。國外也有不少關(guān)于利用轉(zhuǎn)基因大鼠的報(bào)道?紤]到多巴胺受體在大腦功能方面的重要作用,我們利用CaMKⅡ啟動(dòng)子構(gòu)建了前腦特異表達(dá)D5B受體的轉(zhuǎn)基因大鼠。先將D5B基因克隆入含有內(nèi)含子和SV40 polyA尾巴的p265載體,再將p265—D5B片斷克隆入含有CaMKⅡ啟動(dòng)子的p279載體。轉(zhuǎn)基因大鼠通過顯微注射獲得,并用PCR和Southern blot的方法鑒定轉(zhuǎn)基因陽性。
