益生菌制劑生物效應(yīng)的實(shí)驗(yàn)研究

【摘要】: 隨著經(jīng)濟(jì)的飛速發(fā)展和人民生活水平的提高,人們對(duì)生活質(zhì)量和自身健康的要求在不斷提高。自發(fā)現(xiàn)益生菌以來(lái),益生菌對(duì)人們健康有益和有效的研究正越來(lái)越受到重視。目前,益生菌產(chǎn)品正成為大眾消費(fèi)的一部分,以益生菌制劑為核心的生產(chǎn)、消費(fèi)群正變得越來(lái)越大。益生菌這種活性菌群能調(diào)節(jié)腸道內(nèi)原生微生物菌群的平衡,從而有益人體健康。然而,益生菌產(chǎn)品究竟給人們健康帶來(lái)哪些影響,這是大眾關(guān)心所在,同時(shí),也是該領(lǐng)域科研的熱點(diǎn)。 為此,本研究設(shè)計(jì)了一系列的活體動(dòng)物實(shí)驗(yàn),采用了形態(tài)學(xué)觀測(cè),免疫比濁法,模型對(duì)照實(shí)驗(yàn)法,噻唑藍(lán)(MTT)比色分析等方法,考察益生菌制劑對(duì)動(dòng)物消化系統(tǒng)、免疫系統(tǒng)和循環(huán)系統(tǒng)的影響。測(cè)試了實(shí)驗(yàn)小鼠的腸道運(yùn)動(dòng)功能,腸道pH變化,腸壁形態(tài)變化,胸腺系數(shù)、脾臟系數(shù),血清中IgA、IgM、IgG、C3、C4、膽固醇、甘油三酯、高密度脂蛋白、低密度脂蛋白含量,血液高低切粘度,光密度值等指標(biāo)。 實(shí)驗(yàn)表明飲用菌劑的實(shí)驗(yàn)組與對(duì)照組相比,實(shí)驗(yàn)組小鼠腸道運(yùn)動(dòng)顯著加強(qiáng)(P0.01),腸道粘膜形態(tài)得到改善;脾臟指數(shù)(P0.05)、C3(P0.01)、IgA含量有顯著提高,血清膽固醇含量顯著降低(P0.01)、甘油三酯含量也有所降低(P0.05)、高密度脂蛋白、低密度脂蛋白含量沒(méi)顯著變化,而血液低切粘度有很大的改善,MTT比色實(shí)驗(yàn)所測(cè)光密度值隨試劑濃度增大而減少,細(xì)胞相對(duì)增殖率隨濃度增大而降低,益生菌制劑液能抑制C26細(xì)胞的增殖。 以上實(shí)驗(yàn)結(jié)果分別從消化、吸收、免疫,血液學(xué)等方面,證實(shí)益生菌制劑的確有利于增強(qiáng)動(dòng)物的消化功能,可促進(jìn)機(jī)體免疫抗體球蛋白合成,提高機(jī)體免疫力,減少膽固醇含量,降低血液粘度。實(shí)驗(yàn)結(jié)果可以為當(dāng)前益生菌作用機(jī)理的理論研究提供實(shí)驗(yàn)參考。
[Abstract]:With the rapid development of economy and the improvement of people's living standard, people's demands on quality of life and their own health are increasing. Since the discovery of probiotics, more and more attention has been paid to the beneficial and effective study of probiotics to people's health. At present, probiotic products are becoming a part of mass consumption, the production of probiotics as the core, consumer groups are becoming larger and larger. Probiotics, a group of active bacteria, can regulate the balance of intestinal protozoa, which is beneficial to human health. However, the influence of probiotic products on people's health is the focus of public concern and scientific research in this field. Therefore, a series of animal experiments in vivo were designed in this study. Morphological observation, immune turbidimetry, model control experiment and (MTT) colorimetric analysis were used to investigate the effects of probiotics on the digestive system of animals. The effects of the immune system and the circulatory system. The changes of intestinal motor function, intestinal pH, intestinal wall morphology, thymus coefficient, spleen coefficient, serum IgA,IgM,IgG,C3,C4, cholesterol, triglyceride, high density lipoprotein, low density lipoprotein were measured. Blood viscosity, optical density and other indicators. The results showed that compared with the control group, the intestinal motility of the experimental group was significantly enhanced (P0.01), and the intestinal mucosal morphology was improved. Spleen index (P0.05), C3 (P0.01), IgA), serum cholesterol (P0.01), triglyceride (P0.05), high density lipoprotein (HDL-C) were significantly increased. The low density lipoprotein content did not change significantly, but the low shear viscosity of blood was improved greatly. The optical density value of MTT colorimetric experiment decreased with the increase of reagent concentration, and the relative proliferation rate of cells decreased with the increase of concentration. Probiotics can inhibit the proliferation of C26 cells. From the aspects of digestion, absorption, immunity, hematology and so on, the above results prove that probiotic preparations are indeed beneficial to enhance the digestive function of animals, promote the synthesis of immunoglobulin, and improve the immunity of the body. Reduce cholesterol content, reduce blood viscosity. The experimental results can provide experimental reference for the theoretical study of probiotic action mechanism.
