不同新生隱球菌菌株對人臍靜脈內(nèi)皮細胞損傷的初步研究

【摘要】:目的:探討不同新生隱球菌菌株與人臍靜脈內(nèi)皮細胞(HUVEC)共孵后,新生隱球菌對HUVEC的抑制率和凋亡率的差異,為進一步的蛋白質(zhì)組學篩選最大差異的菌株。方法:通過透射電子顯微鏡觀察新生隱球菌侵入和損傷HUVEC的過程以及作用后細胞的形態(tài)學變化;并分別在不同時相點用CCK—8試劑盒檢測細胞抑制率的變化;用流式細胞儀檢測兩者相互作用后細胞凋亡率的差異。結(jié)果和結(jié)論:通過透射電子顯微鏡觀察到三種新生隱球菌菌株均有出芽狀態(tài),造成細胞不同程度的損傷。CCK—8試劑盒的檢測結(jié)果表明,共孵30分鐘時,三者的抑制率比較無統(tǒng)計學意義,共孵時間分別為1小時,,2小時,3小時,4小時時,莢膜缺陷株和白化株對內(nèi)皮細胞的抑制率均高于野生株,5小時時,三者的抑制率在統(tǒng)計學上無明顯差異。流式細胞儀的檢測結(jié)果與其基本一致。
[Abstract]:Aim: to investigate the difference of inhibition rate and apoptosis rate of Cryptococcus neoformans (Cryptococcus neoformans) on HUVEC after co-incubation with human umbilical vein endothelial cells (HUVEC). Methods: the process of Cryptococcus neoformans invasion and injury of HUVEC and the morphological changes of cells were observed by transmission electron microscope, and the inhibition rate of Cryptococcus neoformans was detected by CCK-8 kit at different time points. Flow cytometry was used to detect the difference of apoptosis rate between the two groups. Results and conclusion: three Cryptococcus neoformans strains were observed to have budding state by transmission electron microscope, resulting in different degree of cell damage. The results of CCK-8 kit showed that after 30 minutes incubation, The inhibition rates of the three strains were not statistically significant. The inhibitory rates of the capsule defect strain and albino strain on endothelial cells were higher than that of the wild strain at 1 h, 2 h, 3 h and 4 h, respectively, and at 5 h, the inhibition rate of the three strains on endothelial cells was higher than that of the wild strain. There was no statistical difference in the inhibition rate among the three groups. The results of flow cytometry were in good agreement with them.
