PKC在CCK-8調(diào)節(jié)RAW264.7細(xì)胞CD14表達(dá)中的作用

【摘要】:目的:單核-巨噬細(xì)胞的活化在誘導(dǎo)炎癥反應(yīng)中起關(guān)鍵作用。內(nèi)毒素的主要活性成分脂多糖(Lipopolysaccharide,LPS)或其它致炎因素激活巨噬細(xì)胞釋放大量炎癥介質(zhì),包括TNF-α、IL-1β和IL-6 等,引起機(jī)體自身組織損傷,導(dǎo)致過(guò)度的全身性炎癥反應(yīng),最終導(dǎo)致多器官功能障礙以至死亡。CD14 是巨噬細(xì)胞識(shí)別LPS 的關(guān)鍵受體,LPS 與LPS結(jié)合蛋白(Lipopolysaccharide binding protein, LBP)形成復(fù)合物后,與CD14 結(jié)合激活巨噬細(xì)胞,因而CD14 是LPS激活細(xì)胞發(fā)揮效應(yīng)的始動(dòng)環(huán)節(jié)。CD14 包括膜結(jié)合型CD14(membrane bound CD14, mCD14)和可溶性CD14(solubleCD14, sCD14)兩種形式。 八肽膽囊收縮素(cholecystokinin octapeptide, CCK-8)具有一定的抗炎及抗休克作用,可明顯減輕內(nèi)毒素休克(endotoxin shock, ES)大鼠肺臟、肝臟、脾臟、腎臟間質(zhì)水腫及白細(xì)胞浸潤(rùn)等炎性病理?yè)p傷。但有關(guān)CCK-8 的抗炎作用及其機(jī)制尚未完全闡明。蛋白激酶C(Protein Kinase C,PKC)作為重要的信息分子廣泛參與機(jī)體免疫調(diào)節(jié)和炎癥反應(yīng)的信號(hào)轉(zhuǎn)導(dǎo)。許多研究證實(shí)PKC 是參與LPS 誘導(dǎo)CD14表達(dá)的重要信號(hào)介質(zhì)。 最近本室研究表明,在肺間質(zhì)巨噬細(xì)胞(pulmonaryinterstitial macrophages,PIMs),CCK-8 可明顯抑制LPS 誘
[Abstract]:Objective: the activation of mononuclear macrophages plays a key role in inducing inflammatory response. Lipopolysaccharide (Lipopolysaccharide,LPS), the main active component of endotoxin, or other inflammatory factors activate macrophages to release a large number of inflammatory mediators, including TNF- 偽, IL-1 尾 and IL-6. CD14 is a key receptor for macrophages to recognize LPS. After forming a complex with LPS binding protein (Lipopolysaccharide binding protein, LBP), LPS binds to CD14 to activate macrophages. Therefore, CD14 is the initiation of LPS activation. CD14 includes membrane binding CD14 (membrane bound CD14, mCD14 and soluble CD14 (solubleCD14, sCD14). Cholecystokinin octapeptide (cholecystokinin octapeptide, CCK-8) has some anti-inflammatory and anti-shock effects, and can obviously alleviate inflammatory pathological damage such as lung, liver, spleen, kidney interstitial edema and leukocyte infiltration in (endotoxin shock, ES) rats with endotoxic shock. However, the anti-inflammatory effect of CCK-8 and its mechanism have not been fully elucidated. As an important signal molecule, protein kinase (C (Protein Kinase) is widely involved in the signal transduction of immune regulation and inflammation. Many studies have confirmed that PKC is an important signal medium involved in CD14 expression induced by LPS. Recent studies in our laboratory have shown that CCK-8 can significantly inhibit the induction of LPS in pulmonary interstitial macrophages (pulmonaryinterstitial macrophages,PIMs).
