乳豬肝細(xì)胞、Kupffer細(xì)胞共培養(yǎng)時(shí)的生長(zhǎng)及功能

【摘要】:目的 建立穩(wěn)定高效分離乳豬肝細(xì)胞與Kupffer細(xì)胞的實(shí)驗(yàn)流程,研究乳豬肝細(xì)胞與Kupffer細(xì)胞共培養(yǎng)時(shí)對(duì)肝細(xì)胞的生長(zhǎng)、代謝、解毒等生理功能的影響,為構(gòu)建高效、經(jīng)濟(jì)實(shí)用的人工肝系統(tǒng)提供參考數(shù)據(jù)。方法 采用Ⅳ型膠原酶離體灌注法分離單個(gè)乳豬肝細(xì)胞,聯(lián)合percoll液密度梯度離心法分離Kupffer細(xì)胞,比較肝細(xì)胞單獨(dú)培養(yǎng)和以不同比例與Kupffer細(xì)胞共同培養(yǎng)時(shí)對(duì)安定和醋氨酚的清除功能和生存時(shí)間。結(jié)果 采用本方法每頭乳豬可分離得到單個(gè)肝實(shí)質(zhì)細(xì)胞數(shù)為(3.5±1.1)×10~9,,活性大于90%±2%。Kupffer細(xì)胞的產(chǎn)量平均為(2.35±0.5)×10~8,純度為95%±4%。肝細(xì)胞與Kupffer細(xì)胞按12∶1比例混合時(shí)較8∶1和16∶1混合時(shí),鏡下細(xì)胞形態(tài)學(xué)上生長(zhǎng)更好,對(duì)安定和醋氨酚的清除顯著優(yōu)于后兩者,并且功能可維持兩周左右;而以16∶1及8∶1混合和肝細(xì)胞單獨(dú)培養(yǎng)時(shí),肝細(xì)胞雖能存活十天以上,但一周左右肝細(xì)胞功能就明顯減低。結(jié)論 1.通過(guò)本實(shí)驗(yàn)研究成功地建立了以離體肝灌注的Ⅳ型膠原酶消化分離乳豬肝實(shí)質(zhì)細(xì)胞及聯(lián)合Percoll液密度梯度離心法分離Kupffer細(xì)胞的實(shí)驗(yàn)室操作流程,該流程穩(wěn)定高效實(shí)用,分離得到的肝細(xì)胞及Kupffer細(xì)胞數(shù)量基本能滿足生物人工肝對(duì)肝細(xì)胞數(shù)量的要求,可用于構(gòu)建生物人工肝。2.按8∶1、12∶1和16∶1的比例在體外進(jìn)行肝實(shí)質(zhì)細(xì)胞與Kupffer細(xì)胞264小時(shí)原代貼壁共同培養(yǎng)實(shí)驗(yàn),實(shí)驗(yàn)結(jié)果提示,肝實(shí)質(zhì)細(xì)胞和Kupffer細(xì)胞在適宜的培養(yǎng)條件下可進(jìn)行大量共培養(yǎng);這兩種肝細(xì)胞的同步貼壁簡(jiǎn)單共培養(yǎng),肝細(xì)胞的生長(zhǎng)、解毒及生理功能將受到影響,并且按12∶1比例混合培養(yǎng)時(shí)較8∶1和16∶1以及肝細(xì)胞單獨(dú)培養(yǎng)生長(zhǎng)及功能上為佳,細(xì)胞生物學(xué)活性也高,生長(zhǎng)也較好,這些數(shù)據(jù)提示我們當(dāng)肝細(xì)胞和枯否細(xì)胞體外共培養(yǎng)時(shí)選擇合適的比例、時(shí)機(jī)和方法以及更適合的培養(yǎng)系統(tǒng)是需要的。我們發(fā)現(xiàn)12∶1比例生長(zhǎng)及功能更好并可用于構(gòu)建生物人工肝。
